GFJ also inhibits CYP2D6 to some extent if I remember correctly. However I used both GFJ and Zoloft (both CYP2D6 inhibitors) with codeine once upon a time and didn't notice that much decrease in potency. But at the same time, I've always been able to go way over the ceiling dose with codeine and get extremely buzzed off 300-400mg without a tolerance.

Alcohol is a CYP2D6 inducer, which was recommended to me a while back on this forum by another user who I can't remember now... they recommended a glass of wine or a beer about 45 minutes or so before dosing codeine. CYP2D6 is what converts codeine to morphine, so having more CYP2D6 working on your codeine is a good thing.

Of course, don't get drunk and then dose your codeine, the combo is very unpleasant IME. And if your drunk there are dangers of CNS depression when adding in any other downer.

Yes, 240mg should be fine for a first timer, my first time with codeine was 12 N+ which made me slightly relaxed and happy. As belarki notes, going above 400mg for most people (not me) is both useless and also will give you the histamine reaction from hell. The LD50 (dose of codeine that will kill 50% of the people who take it) is estimated to be 700mg, and the thing that would most likely kill you would be the overt histamine release.
