When mephadrone first came under legal control we were sent a sample of 3-fluoro-2-(methylamino)-1-(4-methylphenyl)propan-1-one (SMILES CNC(CF)C(=O)c1ccc(C)cc1) and what was interesting was that the compound was more or less identical if consumed orally BUT it stung like crazy if one was dumb enough to try snorting it.

Why?

Because the Chinese manufacturer had simply made the hydrochloride salt. I even asked them if they could produce various different addition salts so we could perform some basic tests. I don't just mean solubility but details such as stability. As soon as you end up with hemi or tripos addition salt, there may be a tendency for the product to be hygroscopic which poses problems of storage and ROAs. It turned out that they had optimized their mephadrone production so much that introducing a different addition salt would have caused them major difficulties. I can only presume they had a devoted production line.

I'm certainly surprised that we have yet to see those examples with longer alkyl chains not simply replacing the terminal methyl moiety for a fluoromethyl moiety. Cost is, I assume, the issue. Amazing to think that pyrophenidone first turned up about a deade ago but it's only now that others are offering it as an RC. It was hardly a work of genius given that lefetamine has been known since the 1940s and that zylofuramine simply underlined the fact that the B-ring is simply a bioisostere of an N-propyl moiety.

Zylofuramine is also interesting because in a similar manner to the cathinones, there is an amine function on a carbon adjascent to a carbon with an oxygen containing function. As far as I can tell, it's the ability of the lone-pairs to interact that makes tertiary amines MORE potent than secondary amines which in there turn are more potent than primary amines.
