When you take something - anything - orally it goes to your stomach, and then small intestine where it then gets absorbed into the bloodstream.... This blood flows directly to the liver where many drugs are metabolised to some degree, before they then go off to the brain where they produce the effects...
This is known as first pass metabolism.
When you plug drugs, they get absorbed into the bloodstream and this blood flow bypasses the liver and heads straight for the brain, thus avoiding any first pass metabolism... Basically getting more bang for your buck.
Take MDMA for example... With an oral dose only about 50% reaches the brain and produces an effect. When you plug the same dose, approx 90% will reach the brain. In effect, by plugging you are double dropping... Plus I believe there is a quicker onset of action...
