Study said:Abstract Purpose. The possibility of using polysorbate 80-coated nanoparticles for the delivery of the water insoluble opioid agonist loperamide across the blood-brain barrier was investigated. The analgesic effect after i.v. injection of the preparations was used to indicate drug transport through this barrier.
Methods. Loperamide was incorporated into PBCA nanoparticles. Drug-containing nanoparticles were coated with polysorbate 80 and injected intravenously into mice. Analgesia was then measured by the tail-flick test.
Results. Intravenous injection of the particulate formulation resulted in a long and significant analgesic effect. A polysorbate 80 loperamide solution induced a much less pronounced and very short analgesia. Uncoated nanoparticles loaded with loperamide were unable to produce analgesia.
Conclusions. Polysorbate 80-coated PBCA nanoparticles loaded with loperamide enabled the transport of loperamide to the brain.
^ That's old news, man. Plus, no one without sophisticated lab equipment and materials will be able to do that.

Yes unfortunately it has TURNED into old news. Im the one who created this thread and really hoped it would not devolve into the old BBB debate (hence my title of this thread)...oh well.
could be psychological. as in: all peripheral symtoms of w/d reversed --> reactions in the brain to the now beautiful signals coming in, "must have taken an opiate" --> psychological relief of central symptomsoff topic here I'm sorry... however, I took loperamide, around 14mg's yesterday (not the disgusting doses we've read about when people try to abuse it) when I ran out of suboxone. As well as keeping diarrhea at bay, I am adamant that it reversed my cold chills and sweats. Simply put, before I took the loperamide I had sweats and cold chills, afterwards they were gone. Now, how could this be if the sub blocks the action of any opiates? I know that loperamide doesn't cross the BBB. So what happened? I am extremely sensitive to opiate wd's and know my body pretty well after 10yrs of opiate addiction so I'm almost 100% this was not a placebo. Did the loperamide absorbed thru my gut relieve the opiate wd's? I was sick of the debate of loperamide myself and didn't think I'd contribute to the posts/misleading/confusion, but I was just too uncomfortable and wanted to try something. I'd have to try it again to be more certain (I have subs again) but yesterday's experience was positive for loperamide.

If you read my first post, you will see that i really did make an honest attempt to discuss this subject from an entirely different angle, but for whatever reason, there has been no discussion along the lines i hoped, and instead this whole thread became loperamized. Well...at least a new term has been invented which might catch on.

I know the difference between placebo and actual effects.
If it were placebo, the unique effects like that wouldn't be there. It would just feel like you took any other opiate since your brain just sees it is taking an opiate. It wouldn't have such specific effects if it was only placebo.
