Misc Route of Administration question

I'm afraid I have no idea. Pregabalin is a relatively new drug and I don't think there is a large body of experience on the ROAs or potentiators. I'm not going to try putting it up my butt though!
You know what im going to try it tomorrow in a paraqchute another low but ive had plenty in my drug life ill let you know how it goes
 
I take vastly more - I'm talking several GRAMS at a time!

I do worry what detrimental effect it might have on my body. According to @MsDiz - who is a doctor, and wrote in the Preg thread in EADD - the main consequences of long term Pregabalin use is cognative decline/dementia
Youy must have noticed how short term memory gets fucked on it i would play a game i love rock led zeppelin and lots other i would try to name members of bands when on pregabalin i would forget my favorite band members also makes me talk alot but i love the buzz
 
Also when i was on huge doses i fucked my kidneys not permanatly but had to take pills. It started with foamy piss then water like feeling on lungs bad joints better now but they also fuck kidneys up at massive doses

damn - did you go to a doctor, and what did they say? Are your kidneys OK now?

I like the buzz too. I guess it's a bit like alcohol in that it imapares your judgement and motor skills. I much prefer Preg to alcohol though - which I can barely stomach any more. Although at least booze doesn't have the rapid tolerance increase like Preg does

My kidneys were fucked something called creatine levels plus something else they gave me pills i been overdoing drugs for 30 years but pregabs fucked me up mentally and physically more in just 2 years of using

I explained it to a mate as having the energy of a coke buzz with no edgyness to it slightly pissed and its weird when you need to be chatty and up you up put your head down and you knocked out and music fuck me music sounds the best it can compared to any drug i done se that great and it gives you the staying power like opium but unlike opium it makes you horny as fuck apart from crank or crack or heroin it my favorite drug

I hope your kidneys are OK now. I guess you didn't tell the GP why your kidneys were fucked, or did you admit the truth?

They good now brother and fuck no i never tell the doc anything about my drug use nothing good comes from it i did try to get some prescribed gave symptoms of fibomyalgia he told me to take ibuprofen the cunt

One thing about taking a large quantity of Pregabalin, my family notice the next day that I have noticable droopy eyes - something I can't hide

Preg can make me feel great, but there's always a catch - and in this case it's the rapid tolerance increase. Annoying, but probably a good thing for Harm Reduction purposes
 
As other people have stated it depends on the substance. If you want to check for specific substances and roa look up bioavailability of the specific substance from the different roa. Easily found in references like TPDR.

Thanks I've looked at the TPDR site and it says:

Oral Route
Oral bioavailability of pregabalin is roughly 90% and is independent of dose. Oral formulations are rapidly absorbed. The immediate-release oral solution and capsules are bioequivalent dissolving at a rate of more than 85% within 30 minutes. Pregabalin displays linear pharmacokinetics. The median Tmax for immediate-release formulations is 0.7 hours (0.7 to 1.5 hours), and for extended-release tablets is 8 hours (5 to 12 hours). After multiple oral doses, steady state concentrations are achieved within 1 to 2 days for immediate-release formulations and 2 to 3 days for extended-release formulations. Food delays the rate but not the extent of absorption of immediate-release formulations. Administration of the extended-release tablets once daily after an evening meal has an equivalent AUC and lower Cmax compared to administration of immediate-release formulations without food twice daily.

so it seems like oral is the best way for Pregabalin at 90% bioavailability, but of course this site isn't going to discuss the effectiveness of other ROAs

The PDR might not, but it's worth searching PubMed for "pregabalin + sublingual" and so on.

Of course they don't care about helping us out. But coming up with a new mechanism for delivering a drug is one of the ways Big Pharma can "evergreen" products that have gone generic, as Lyrica has in the US recently.

They get a new patent for an orally dispersing tablet or an extended release form or a nasal spray or a sublingual film.

So they're very interested in funding research on ROAs. It's also relatively easy to conduct studies on pharmacokinetics, since they can be done with a small sample group of non-patients.

Pregabalin goes through little to no metabolisation with something like 97% being excreted unchanged in urine.

@yubacity there are rare cases that I’ve seen where gabapentin has done reversible damage to the kidneys.

Pregabalin is still a funny drug that’s being researched in regards to side effects etc but for those with issues with their kidneys as I’m sure you probably may have had due to your use of other drugs then it’s not advisable to use it.

@Forever Changes just to say, whilst I trained as a Dr and practiced for a year I moved straight into research science. I wouldn’t and can’t give advice from the standpoint of being a Dr. I just try to explain what I know from my observations and my own prescribed use due to my own health issues.

Edit to add: whilst I can give very a general explanation it in no way makes up for what your own Dr can tell you. They know your medical history etc and your first port of call especially in regards to kidney issues should always be your own Dr.

@MsDiz - I totally understand, I know you can't and shouldn't give medical advice out to people here who you don't know aren't your patients, but I do value your advice

so if 97% of Pregabalin is excreted unchanged then only 3% of what you ingest is metabolised and used by your body? But of that 3%, 90% is bioavailable when you take it orally? Have I got that right?

Seems like an awful waste if only 3% of a capsule is actually used by your body.

But if taken sublingually, it goes straight into your bloodstream - so might that be more efficient, or is it necessary to go through the gastric system/liver to be metabolised properly?

Could be mate i heard that before but that the only new drug added might just be my kidneys were going to get fucked anyway . Not that i could ever do this does that mean if i drank my piss i could keep recycling pregabalin with only topping up sometimes

For someone with kidney issues the last thing you want to be doing is drinking your own piss.

It works a bit differently. Bioavailability (here 90% for oral) is how much of substance enters your bloodstream. With IV it's always 100%. This now passes the liver (what snorting circumvents) where some drugs are being metabolized (enzymatically transformed into another molecule, active or inactive) and then stuff reaches the brain. Then again liver, next part gets metabolized etc until down to zero. Drugs don't get used up by the body, they act rather like keys in a lock.

This is the usual case. Now with pregabalin only 3% getting metabolized means more of it stays around for longer and can occupy more receptors - this is part of why pregab is more potent than gabapentin. Other is it gets actively transported over the blood brain barrier by some pump.

How the body excrets stuff is another chapter I still have to learn.

But with pregabalin you get no real plus by using other RoA's. With e.g. morphine (poor bioavailability) or ketamine (pronounced first pass = metabolized before reaching the 🧠 ) things are different.
 
For someone with kidney issues the last thing you want to be doing is drinking your own piss.
Never would was tempted to drink it as a 15 year old with crank my tweaker mate used to say to me i wasting a great by flushing it down the toilet i drew the line. Ok i did not draw the line my tweaker piss was orange and smelled really bad i was gagging with the smell
