Yes you will. Parts of the tolerance will be permanent i.e. the feeling that you've found "an answer" and total euphoria. Mood lift, anorexic effects, stimulation, and to a certain extent empathy can be prolonged by taking tolerance breaks. Negative effects such as irritability and perhaps depression will become more prominent until you stabilize (I think this can take a few weeks to a few months but I was going to ask a similar question so I might make another thread for this). High blood pressure and effects on concentration and a mild antidepressant effect will moat likely continue indefinitely.
Tolerance to stimulants tends to be hard to get rid of. It could take years of not being on it after taking it for a few months to relive ones first experience. Or it may not be feasible.

I was going to drink after taking one, last time I did some was 4 months ago I had 3 in a week during exams. I've read drinking with them can be fun but I was wondering if I should take one a week before to get use to the feeling or should I just keep my tolerance 0 for best effects/experience?
Well tolerance builds at a medium speed and some aspects never go away. The strictly stimulant effect can be prolonged.
Yes; that depends on your dosage, but considering you're considering taking a more-ish combo its bound to be bad. Combining methylphenidate with alcohol is risky to say the least; you won't be able to tell how drunk you are and it puts undo stress on your heart. No it wouldn't at all.
