Opioids The binding affinity of buprenorphine vs. other opiates...

Okay, so we all know that buprenorphine has a higher binding affinity than most other opiates/opioids, but my question for everyone is what commonly prescribed opiates/opioids (or street drugs for that matter) have an equal or higher B.A. than bupe? Are there any commonly known drugs with a higher B.A., or is bupe pretty much at the top of the list?

I would imagine that the bentley compounds have a higher binding affinity than bupe. I highly doubt any street drugs do, though.

One thing to note is that tramadol and it seems, odesmethyltramadol both synergize with bupe to create a stronger analgesic effect.


We will let this go in od and maybe move it to add after a bit. I will see if sekio has any advice.
 
Interesting question. See page 103 of the book here ( scroll down a page ) for a table of binding affinities at all three major receptors in guinea pig brains at least. Bup is beaten only by Lofentanil for binding affinity across all three receptors if I've read it right.
 
Etorphine (a Bentley compound like muvolution said), probably has a higher binding affinity than buprenorphine. I imagine that there are fentanyl analogues that have a stronger binding affinity as well. However none of these are commonly prescribed. If you're looking for the opioids prescribed in the US that have the best chance of shooting through bupe, they would be fentanyl, hydromorphone, oxymorphone and methadone.

There are a lot of opioid chemicals I'm sure that have a higher binding affinity, but if you ever did manage to somehow come across them, they would most likely be so dangerously potent that using them would be incredibly risky, to say the least.
 
^^ Well, I cannot speak on behalf of fentanyl or oxymorphone having a higher binding affinity than bupe because even though I have used both, it was never while having used bupe beforehand. Hydromorphone on the other hand I am prescribed and I can assure you from many, many tries, it will not have any effect (on me at least) until at least 24 hours after my last bupe shot. Just my $0.02. :)

Anyone else have any input on this topic???

Yeah, oxymorphone doesn't, but it is strong enough to compete. I've blasted through 4mg/day buprenorphine with 5mg IV shot of opana, but then again, 4mg is hardly a blocking dosage. Last spring I was taking 16mg of suboxone a day, and I shot one bag of heroin and it completely broke through the bupe, I got an intense rush followed by a long high that was not diminished at all. It was really strange, and I wonder if that could have even been heroin, or if it was cut with fentanyl or something..
