Sequence of Opiod Dosing with CYP2D6 Inhibitors

First off let me explain the situation: tonight I will be ingesting 100 mg of insufflated tramadol along with my typical hydrocodone-potentiator cocktail.

Question One- Will using cyproheptadine be a useful potentiator of my opiods? Ive never tried this one before and am not sure if it will be effective. I also have chlorpheniramine on hand if that would work instead. I have dimenhydrinate on stand by just in case. Would prochlorperazine effect opioid effects? It's always been good for its anti-emetic effects but it's a neuroleptic correct?

Question Two- Which opioid should I ingest first? I am not sure if the CYP2D6 inhibition will substantially reduce the effectiveness of the tramadol? Would I have to take the tramadol before taking the cyproheptadine or antihistamine give a better effect?

Just to clarify- I will be taking 20 mgs of cyclobenzeprine right before my peak to help the nod. If their is a contraindication with the cyproheptadine, you should let me know. I plan to dose very soon so a little help would be awesome before I go for it.

i think a cyp2d6 inhibitor would really mess up the metabolism of tramadol into o-desmethyltramadol, so in that case i would not bother.
 
I'll only be taking 100 mg that I plan to insufflate. But the dose of hydrocodone will be around 60-70mg so I want to know in what order I should ingest them. Possibly insufflate the tramadol shortly before taking the potentiators? It will draw the high out though.

And the potentiation questions are really bugging me, moreso than the tramadol vs hydro dosing actually.

Edit: I blew the tramadol about 1:15 ago, and took my game potentiation package. I shall update soon: I decided to take 6mg of cyproheptadine, 75mg dimenhydrinate, and 38.5 mg diphenhydramine with .75 mg of clonazepam.

If anyone has used cyproheptadine as an opioid potentiator i'd love to hear the experience.

But the chlorpromazine is a viable option as a potentiator in the future?

I know it's stupid but I just enjoy the feeling much more with I snort tr tramadol. I can't get much out of it orally and I only had two left. Id rather not be judged, I've been on this forum long enough to know the general opinion on tramadol.
 
yeah sorry missed your question, i would have taken the tramadol first, let it metabolize then continue with your regular cocktail about an hour or so afterwards, guess it's too late now. not sure about anything else. Well since you've done it, how did everything work out potentiation wise?
 
I actually did snort the tramadol a little over an hour before I began my potentiation combo. So I think I actually did it the right way oddly enough. I can't say for sure how the potentiation is gonna go so far, I just now dosed (15 mins) 50mg of hydrocodone and 10mg's of cyclobenzeprine so effects will be reported shortly. I am feeling a nice buzz coming on though, with a distant tramadol flare. Maybe O-desmethyl metabolization isn't much of a factor at low doses?

My potentiation cocktail tonight was the same as listed about but I also added 12mg chlorpheniramine and 100 mg of butalbital as well. Keep in mind that I have an incredibly high tolerance to GABAergics though.

Initial update on potentiators: the cyproheptadine and CPM are potentiation better than diphenhydramine or promethazine ever have. I think the cyproheptadine has a lot of potential for potentiation :)

Wow, the new potentiation combo and early dosing of tramadol before aforementioned combo are really putting in some heavy lifting. My buzz is almost at a nod, and I'm still under 60mgs, and my chronic pain is almost completely gone for the time being. I'm not sure but i think the cyproheptadine is largely responsible for this.

Has anyone else used it in conjunction with opioids before?
 
I presume neuroleptics would add sedation, aiding the nod - but IMO anti-psychs just blunt anything, I'm sure the physical side would be bumped up, but the euphoria would be reduced, I could be wrong though.
