Experimental Procedure
  1. Reaction Mixture Preparation: In a dry three-necked flask equipped with a reflux condenser and dropping funnel, dissolve allylbenzene (e.g., 10 mmol) in a suitable, non-polar solvent like benzene or hexane (20-30 mL).
  2. Initiator Addition: Add a catalytic amount of radical initiator, such as benzoyl peroxide (0.1–0.5 mmol), to the solution.
  3. Hydrobromination: While stirring, add a solution of hydrogen bromide (HBr) in acetic acid or bubble gaseous HBr through the reaction mixture.
  4. Heating/Initiation: Heat the mixture to reflux (typically 60–80°C) for several hours. The reaction requires heat or light to initiate the homolytic cleavage of the peroxides.
  5. Workup: Cool the mixture to room temperature. Wash the organic layer with water, followed by a dilute sodium bicarbonate solution

    to neutralize any remaining acid.
  6. Purification: Dry the organic layer over anhydrous magnesium sulfate (Epsom salts), filter, and remove the solvent under reduced pressure using a rotary evaporator. The product can be further purified via distillation or column chromatography.

The nucleophilic substitution of 2-bromopropane with methylamine (an amine alkylation) typically proceeds via an Sn2 reaction mechanism to produce

-isopropylmethylamine.

Because secondary alkyl halides are moderately hindered and amines can react multiple times, this reaction usually requires heat and an excess of methylamine to prevent the formation of tertiary amines.

Experimental Procedure (General Laboratory Scale)
This procedure aims to form the secondary amine, requiring an excess of methylamine to limit over-alkylation to a tertiary amine.
1. Reaction Setup
  • Prepare a round-bottom flask equipped with a magnetic stir bar and a reflux condenser.
  • Add a large excess of methylamine (typically as a 40% aqueous solution or in ethanol) to the flask.
  • Add 2-bromopropane slowly to the methylamine solution while stirring.
2. Heating (Reflux)
  • Heat the mixture under reflux (approx. 50–70°C, depending on solvent) for several hours (often 2–6 hours).
  • The high concentration of methylamine ensures it acts as both the nucleophile and the base to neutralize the
    HBr formed.
3. Work-up and Purification
  • Cool the reaction mixture to room temperature.
  • The resulting solution contains the product (as its hydrobromide salt), excess methylamine, and 2-bromopropane.
  • Make the mixture strongly alkaline (e.g., add 50% NaOH Red Devil Lye Solution) to convert the amine salt into its free base.
  • Extract the product with an organic solvent like diethyl ether or dichloromethane (DCM).
  • Dry the organic layer with an anhydrous drying agent (e.g., MgSO4 or Na2SO4)).
  • Remove the solvent via rotary evaporation.
  • Purify the resulting

    -isopropylmethylamine using fractional distillation.

Key Considerations
  • Over-alkylation: The primary amine product (

    -isopropylmethylamine) is often more nucleophilic than the original methylamine, leading to over-alkylation and the formation of a tertiary amine,

    -diisopropylmethylamine. An excess of methylamine minimizes this.
  • Elimination Competition: 2-Bromopropane can also undergo E2 elimination to form propene, especially with strong bases. However, amine nucleophiles are generally good at substitution.
  • Solvent: Ethanol or methanol is typically used to ensure both the alkyl halide and the amine are soluble.
