Questions on Vyvanse metabolizing

I've been prescribed vyvanse for quite a while now and I was prescribed adderall before that. I know the two are different in the sense that vyvanse MUST be ingested in order for it to take effect because it's a pro-drug. I was wondering if there is anyway to metabolize the pro-drug (lisdextroamphetamine) outside of your system so that you are then left with dextroamphetamine. I know it'd be a lot easier for me to just get back on adderall through my doctor but I just recently had my refill so I'd have to wait quite a while, and I actually need my meds for my ADD and ADHD. I've seen on the forums ideas that include cleaving with amino acids and trypsin, but there is just too much non-specifics for me to understand clearly. I also understand that it is against forum rules to post that in a thread so if anyone does have possible ideas please PM me your ideas, and also possible proof to back up the idea. I of course will accept any idea with or without any sort of documentation back up.

I was also wondering exactly what receptors those amphetamines triggered, such as serotonin or dopamine. This is because I'm wondering if use of my meds could be interfering with my recreational use of MDMA at times by possibly depleting the amount of serotonin in my system.

Amphetamine works with both dopamine and norepinephrine. Serotonin is involved as well, but not nearly as much as dopamine and norepinephrine. Because MDMA works with these chemicals as well, your amphetamine use could be reducing the effects of MDMA, assuming you're using amphetamines often, which it sounds like you are.

If you think something is against the rules why are you asking somebody to PM you? The staff can read PMs too.
 
I'm not sure the specific receptors being hit by D-Amp ,adrenal receptors-obviously, but I do know that D-Amp does not induce anything more than a minimal increase in serotonin bobbing around in the synapses. This is because the serotonin receptors that D-Amp is able to key into are the first casualties of amphetamine-related cell apoptosis and excitotocity - remember the first time you ever tried amphetamine, that awesome feeling never to be seen again? Methamphetamine and MDMA are the amphets with the strong sertonergic profile. But anyway to answer your question, you can build tolerance to amphetamines but I do not think it has anything to do with serotonin depletion/deficiency. The answer probably lies in that as you have slowly built up an amphetamine tolerance, it takes a long time to reset and MDMA being an amphetamine is affected by your tolerance level.
 
Dextroamphetamine is a substrate analog at monoamine transporters. Dextroamphetamine prevents the re-uptake of these neurotransmitters by competing with endogenous monoamines for uptake.

Dextroamphetamine administration increases the activity of the phosphoinositol cycle via an indirect release of Dopamine and Norepinephrine

Transporter inhibition causes monoamines to remain in the synaptic cleft for a prolonged period (amphetamine inhibits monoamine reuptake in rats with a Norepinephrine to Dopamine ratio (NE : DA) of 1:1
At higher doses, Dextroamphetamine can trigger direct release of Norepinephrine and Dopamine. Dextroamphetamine will cause Norepinephrine and Dopamine efflux via transporter proteins, reversing transporter action
Dextroamphetamine releases monoamines in rats with selectivity ratios of about NE : DA = 1:3.5
Serotonin also plays a role in Dextroamphetamine's effect on glutamate levels. However, at therapeutic doses, Dextroamphetamine has meager effect on Serotonin

Yet that is only half of the question I asked. Let's not get anyone in trouble and go about communicating on the matter of metabolizing vyvanse off-site. If you know how to do what I asked of in my main post PM that you can help and then we'll follow up off-site.
