question about dopamine

So I have these adderall. And I know that works by blocking re-uptake of dopamine right?
So then i have these parlodel pills, BROMOCRIPTINE MESYLATE. "direct stimulating effect on dopamine type 2 receptors in the corpus striatum"
i dunno, has anyone else heard of this? i couldnt figure out whether blocking reuptake and then releasing a bunch of it would be too much, or really cool. thanks
 
B]most important[/B] I can not confirm or deny your information. Assuming it is correct (and if it's not the misfortune that results could vary between no high to death), the following applies:
Think of it this way. You're simultaneously preventing the reuptake (absorbtion of excess) and flooding a system. Mmmmmmm.
But check your info and other side effects of the two medicines first. Also you might try looking up the contraindictions of both things you have.
 
I doubt its going to be fun. First of all dopamine has positive and negative effects depending on where administered in the brain. A web search quickly reveals that the dopanergenic system in the corpus striatum is thought to control the motor system (i.e. this will probably affect coordination dizziness and shit like this not produce the wanted high). Moreover, the side effects of this medication include fatigue so it is highly doubtful it will produce the effect you are looking for. In fact I believe you want your dopamine in the nucleas accumbens for the positive effect.
Secondly this page seems to suggest it is the D1 receptors involved in reward behavior. Also as buspar (the antianxiety agent) is a D2 antagonist and does not seem to cause the negative effects one would postulate if D2 stimulation really caused significant reward.
Several other papers however did suggest that D2 was significant in self administrion. So in the end it would be an interesting...if perhaps slightly dangerous experiment. My guess would be that it would at best magnify the effects of this BROMOCRIPTINE shit which doesn't sound abusable.
 
Adderall does NOT cause dopamine reuptake inhibition! Adderall causes a RELEASE of dopamine and norepinephrine.
As for combining Adderall with that second drug, honestly I don't think it would do much. Do the adderall on its own -- it's pretty fun as is.
 
not true..Adderall is a mixed amphetamine (d-amphetamine saccharate, d,l-amphetamine aspartate, d,l- amphetamine sulphate, d-amphetamine sulphate) and all amphetamines have reuptake ability based on potency.
low doses of amphetamine will stimulate the release of norepinephrine, and block the re-uptake of norepinephrine.
moderate doses will stimulate the release and block the re-uptake of both norepinephrine and dopamine
and high doses will effect serotonin
 
DOPAMINE != HAPPINESS
I cannot emphasize this enough! There are around a dozen different pathways of dopamine-using neurons in the brain; while a number of these seem to be involved in some sort of expectation/reward system, a number are not. Even in this reward system, it's a far cry from dopamine=pleasure -- there's a relation between the function of dopaminergic neurons and expectation/learning of reward, but it's not nearly that simple.
Now, the corpus striatum refers to a portion of the limbic system of the brain, mainly the caudate and putamen. Two of the most-studied (and prob important) pathways of dopamine-using neurons converge on the corpus striatum. First, neurons from the substantia nigra, involved mainly in motor movements (this is what quale was talking about) connect to the dorsal (towards the back) parts of the striatum. Neurons from the VTA, involved in expectation/reward/learning (these are the fun ones) connect to the ventral (forward) parts of the striatum, especially the nucleus accumbens. So your drug could be acting on both these systems.
The D2 receptor is indeed intimately involved in the reward functions and is the subject of much study.
There seems to be a misconception that amphetamine doesn't block transmitter reuptake; it does. In fact chemicals that induce release and enter the cell via the reuptake transporter block reuptake to some extent, merely by competition for transporter sites. This is probably the only way in which amphetamine blocks reuptake. [1] contains a detailed discussion of this.
The primary mechanism of action of amphetamine is its direct stimulation of NE and DA release, probably by reversing the reuptake transporter, the same way MDMA works. This is called "carrier-mediated reverse transport." Amphetamine is active as a serotonin releaser, but is very weak. The effective concentrations are below[2].
NE release: 7 +/-1 nM
DA release: 25 +/-4 nM
SE release: 1750 +/-100 nM

Oh sorry, xex, to answer your question -- is the bromocriptine fun on its own? If it is, it'll probably help out the Adderall, I would guess. If it's shitty, then Adderall would prob just increase its shittiness. Sorry I can't offer more info.

I know that cocaine is also a drug that works by blocking the uptake of dopamine. Without sounding stupid, wouldn't it seem that similar effects would occur taking ecstasy and this drug as ecstasy and coke, so the result would be basically no high?
 
of course, there isn't lot that is more beautiful than hallucinations from blingin' after a few days without sleep on glass then taking ecstasy - at least for me! i don't know, i think i fried my brain a while back.

Actually, I think this is a misnomer. Current evidence points to this system as being more focused on expectation/anticipation/learning of reward, rather than actual reward itself. But anyways... I digress...
