Pregabalin has a metabolite called N-methylpregabalin but it undergoes next to no metabolisation. I’ve read studies where 97% of pregabalin is excreted unchanged in urine. You know way more about this than I do Diz, so is pregabalin a prodrug? I.e. does it need to be metabolised to an active compound before it has an effect?
I used to jump in at 1200mg and got royally fucked up, but I've heard that frequent smaller doses are more effective than a large initial dose.
It has something like 90% bioavailability.
Unlike gabapentin which is transported solely by LAT1 (which gets saturated quickly, hence needing to space out doses) pregabalin is also transported by other carriers meaning no saturation of absorption. Which means you really don’t need to space out doses.
All very boring.
