When inoculating the substrate, do you inject through the tape, or remove it? Also, after inoculating, do you put fresh tape over the holes? I am new at this, so please bear with me.

You inject through the tape and immediately put another new one over the tiny hole, or better yet you make a self-healing injection site with something like silicone.

I do the first thing and it's fine.
Removing the tape would probably make a bigger hole for a longer time and may mean that you touch and contaminate things.

If you have a sterile environmentit it shouldn't be a problem to remove the tape and quickly replace it with micropore tape.

The reason to use micropore tape is to letting the substrate breathe while keeping it free of kontamination. You can of course use alternatives to micropore tape.

I have used this method on many substrates and only had very few kontaminations.
