Opiate/amphetamine cross tolerance?

Hey everyone,

Just wanted to know if there was any cross tolerance between Dexedrine taken daily and usage of Oxycontin a day after(not combined with dex)

I know they are very different drugs but they both re-uptake more of the neurotransmitter dopamine to the synapse correct? so would this mean there could be a certain cross tolerance.

I know there is different dopamine sites in the brain which could be activated differently from amphs or oxy, but I just wanted to ask because I found no info on this topic.
 
Always figured Amp in time fried the synapses. I really believe the tolerance for amp is less than percieved. And is more of a compulsion to chase the initial rush, like a drawn out crack buzz. And in fact speed freaks would feel better by holding back, but that is like eating one potato chip. Whereas opiates, it really does take more and more with continued use? Together? Can't say. It would seem the brain to favor a cool groove of opiates to the psychological chaos of amphetamines. But have noticed, used simultaneously, a mild d-amp dose completely buries a comparatively heavy opioid dose. The amount that would normally have a person puking their guts out, barely noticeable.
What this means in synergistic effect on the brain, pharmacologically, pretty curious about that myself...
 
This seems like a question for the ADD dudes. In my experience, taken together, amphetamines and opiates seem to cancel each other out somewhat. The opiate high is negated by the amphetamines and vice versa. I've ended up taking way more than my usual dose of both when using them together. However, a slightly higher dose than normal of opiates with a moderate dose of amphetamines seems to mix quite nicely, but it's probably the safest to start with the opiates and dose the amphetamines a little later.

As far as cross tolerance goes, I'm not qualified to answer.

I have read studies that found there was no cross tolerance between morphine and dextroamphetamine. However, less is known about oxycodone than opiates like morphine - there isn't even agreement about which opioid receptors oxycodone acts on.


It's not fully understood how opioids affect dopamine. Studies have concluded that opioids excite dopamine neurons by hyperpolarization of local interneurons, reducing the spontaneous GABA-mediated synaptic input to the dopamine cells, leading to excitation of the dopamine cells by disinhibition.

Studies have shown that, in select regions, amphetamines increase the concentrations of dopamine in the synaptic cleft, thereby heightening the response of the post-synaptic neuron. There are 2 main theories about the mechanisms by which amphetamines affect dopamine concentrations (which could also both be true at the same time). One theory is that amphetamines acts on the vesicular level, increasing concentrations of dopamine in the cytosol of the pre-synaptic neuron. The other is that amphetamines may interact with the dopamine transporter DAT to induce reverse transport of dopamine from the presynaptic neuron into the synaptic cleft.

So, if these mechanisms of action are correct, it sounds to me like basically amphetamines increase dopamine levels whereas opioids just make the effects of dopamine stronger. Is that correct?

^Yeah, I must have missed it....But does using one drug make you more tolerant to the other drug? I've never heard of that...So, anyone, anyone?
 
^Many drugs can make you more tolerant to other drugs, but it's usually drugs within the same class or with similar effects. For example using one morphine will give you a tolerance to oxycodone, or using psilocybin has a cross-tolerance to LSD, or dextroamphetamine will give you a tolerance to cocaine. Of course cross-tolerance is not always complete and one should never assume there will be a high cross tolerance and therefore start with a large dose when trying a similar drug to one you have a tolerance for. As for dextroamphetamine having a cross tolerance to oxycodone, I personally doubt it.
 
^sorry, yeah....Of course using one opiate would probably make you cross-tolerant to another, the same with amphetamines etc. But I've never heard of drugs in completely different classes causing tolerance to another...If I had to assume, maybe barbiturates and benzos, or like you said, cocaine and amphetamines...But, opiates and amphetamines seems like a stretch...Although, maybe we're wrong...
