Synthetix' post above aside, which I don't doubt but think is a most unusual reaction, assuming you don't suffer withdrawal symptoms from other opiates you might be using you're very unlikely to suffer any withdrawal symptoms whatsoever after a single use, no. The danger with methadone is the long half-life. Depending on your metabolism half-life could be anywhere between 15 and 60 odd hours, call it around 30 hours on average, so if you got into the habit of use for a day, take 2-3 days off, use for a day, take another short break, if you're dosing high enough you could continually have enough opiates in your system to be surfing a habit within a coupla weeks of it.
It's a pretty shitty buzz. Not especially recreational IME, having none of the usual opiate euphoria being pretty much pure sedation. Dose too high good chance you'll be projectile vomiting sick as a pig for a coupla days too. Not especially pleasant stuff. Remember the long half-life also means subsequent daily doses will be cumulative. Take 10 mg today, then another 10mg tomorrow for example and you're gonna have 15mg+ in your system. That way lies overdose territory. Treat with caution.
