MDMA & Oxytocin

^^^^

i was not indicating toxicity per se, but just an alteration/impairment in the effectiveness of the neurotransmitter system of enough nature to inhibit said effect from manifesting

Possibly (again this is just me speculating) MDMAs effects on oxytocin are the first ones to quickly build tolerance.
afaik oxytocin receptors are working in opposite way, the more they get — the more sensitive they are

^Interesting. Do you have a link?
you'll laugh at me, but I found it here — reuniting.info|science|sex_in_the_brain
replace | with slashes,"You are only allowed to post images and URLs to other sites after you have made 10 posts or more." :(

The more oxytocin you produce, the more receptive you are to it. This is the opposite of dopamine. Addicts need more and more of a drug, which, of course, actually means they need more and more dopamine. Luckily you don’t need an ever-increasing "fix" of oxytocin to maintain the same gushy feeling. In fact, your partner just looks better and better - at least to you. This is why this practice can strengthen your bond with your mate.

yes I am sceptical about the source too :D however they also wrote that meditation produce more oxytocin so it looks quite logical to me

I posted a thread about oxytocin agonists a while back.

Check it out.

i'm quite sure oxytocin is at least partly responsible for the the "magic" of MDMA. Hopefully when tolerance develops it's due to the 5ht2 receptors (which are responsible for MDMA induced oxytocin release) and not the actual oxytocin receptors themselves.

And I also think that the ridiculous flood of serotonin might not be fully responsible for MDMA's psychedelic effects. I think MDMA might bind to the 5-HT2A receptors as well at a somewhat low affinity. I read this one report where this guy combined an MAOI and a single E pill and almost died from serotonin syndrome. He experienced heavy hallucinations though which isn't typical of normal serotonin syndrome even at that level of strength. It seems to me that MDA and MDMA stimulate the 5-HT2A receptors, with MDA doing it somewhat more so. It's not that far out either since mescaline and the 2C's are also phenethylamines and are of course, psychedelic.

See this thread: http://www.bluelight.ru/vb/showthread.php?t=209952

MDMA should not have the ability to moderate psychedelic sorts of effects itself with its affinity for 5HT2, however it's important to remember that a fair amount of MDMA is demethylated to MDA, of which one stereoisomer does have a profound effect upon 5HT2. I think this would be related to the psychotomimetic effect of MDMA; an experiment would be to apply something to inhibit n-demethylation. Ketoconazole and ellipticine are some such inhibitors (the first probably being the safest and most well studied), however whether the specific effect would confer to MDMA n-demethylation is unknown.
