What is the dose to maximize norbup accumulation, less than a mg, 1mg, 2mg . Is it better to dose once daily, or multiple times daily to produce most norbupre. What is the ceiling dose where norbup has higher concentration than bup? 4mg? Some say 2, some say 4mg. Is it better to inhibit or induce hepatic enzymes for maximal norbupre production. Like better to take saint John's wort to induce enzymes or tagamet to inhibit for.the most norbup to.be metabolized

I'm afraid I'm not going to be able to answer your question about enzymes to the ability that someone else here will, but I can help with the others. As far as dosing goes (obviously everyone is different, but) 4mg seems to be the generally accepted dose where bupe has all of your receptor sites saturated (thus leaving none open for bupe's beautiful metabolite, norbupe). And although bupe's antagonistic effects can last up to 36+hrs, it's analgesic effects only durate in the 6 hour range. My estimation (coming from both knowledge of the compound and extended experience with changing my dose, ROA, etc.) is that your ideal dose is going to be TOPS 2mg, re-dosing every 6-8 hours or so. However, let me also be the first to tell you that if you're on maintenance (have tolerance), norbupe's greater presence will be hardly detectable until you've tapered down to that being your regular dose anyways.

I'm sure it depends what ROA you use.

When I take buprenorphine, I'm not even thinking about the metabolite or how to maximize it. I get good effects from buprenorphine as is, and I enjoy the effects for what its worth, and I don't look to get anything more out of it.
