Misc Lyrica's Mechanism of Action?

Well I know that Lyrica (pregabalin) is a GABA agonist. But how does it work exactly? Does it work on GABA-A and GABA-B receptor sites? If I go back on Lyrica again, is it going to effect when I use GHB?

I won't be using both at the same time.. but is Lyrica going to down regulate my GABA receptors?

Any info would be appreciated on pregabalin's MOA.

It's not a GABA agonist, I thought it increased GABA levels by increasing the amount of the precursor necessary for that neurotransmitter in the brain. I imagine it would then cause down regulation of your GABA receptors.
 
^ I guess you could call it that... but yes it will down regulate the receptors. Not to the extent of benzodiazepines though I imagine.

Be careful combining with GHB as there will be increased CNS depression.
 
I don't know it's exact mechanism but it's not like how a benzo works. It will down regulate GABA, since your brain needs less of it now you're taking a med, but again I'd say much safer than a benzo / GHB.

This doesn't mean much to me, but quick research on the net tells me it combines with the calcium mechanisms in the body via the spinal cord. I think various other anti seizure / epilepsy drug work that way too.

However u look at it, it's MY wonder drug. I believe it saved me a little, but I don't think I'll be getting any more. I'm tapered right down so maybe it's time to jump ship and see how I go.
 
Lyrica binds to the alpha2-delta auxiliary subunit of voltage-gated calcium channels. Blockade inhibits the calcium-dependent release of a number of neurotransmitters. The drug is a structural derivative of GABA, though it does not bind directly to GABAa, GABAb, or benzodiazepine receptors, does not augment GABAa responses in cultured neurons or have acute effects on GABA uptake or degradation.
 
sorry to bump and old thread but it was a disscussion of lyrica's pharmacology i read somewhere that it acts on the ghb receptor complex somehow but i cannot recall the exact mechanism or source (or i could be tripping) but im fairly sure it did somehow as it has paradoxical stimulant effects. wondering if anyone had anymore insight into this?
