Fluconazole and Oxycodone

Fluconazole is an antibiotic for some female problems but is is CYP3A4 inhibitor which is the main enzyme for metabolizing oxycodone through desmethylation into inactive metabolite.

Ketoconazole has been studied with oxy and it gave 2 to 3 fold rise in AUC.

I guess those are quite similar in inhibition power so I am asking if I should take my normal dose of oxycodone after taking fluconazole or should I decrease it?

2 to 3 fold rise in AUC could mean that it is stronger or lasts longer or both so I am just asking if someone has experiences or insight.

I'd just stick with you can always take more, but you can't take less approach for the future. Good for you for doing your research about interactions! How did it go?

I dont believe fluconazole is quite as strong as ketoconazole at 3A4 inhibition but am going to double check because Im drawing a blank on where I recently read info regarding fluconazole and interactions. I shall return.

Papers arent the easiest but both show fluconazole not as powerful an inhibitor compared to many other azole antifungals including ketoconazole. (And one uses opioids including oxy).

FDA says both ketoconazole and intraconazole are classified as strong 3A4/5 inhibitors with AUC increases >5x while fluconazole is only moderate with AUC increases between 2x and 5x.
