I have been afraid of roller coasters my whole life. I was wondering if a certain drug would change my mindset while on it that would actually make me desire to want to ride on one. Not that I would take something simply to do this. Just wondering if any drugs had fear cures or something.

GABAergics like benzos, GHB, or ethanol all have properties that remove inhibitions which could lead to a person being less afraid of and more willing to do something that scares them. They also relieve anxiety so.... I'd personally go with a good anxiolytic benzo, if I had to choose!

No. No drug is going to change your fears or desires without work. Certain GABA-acting drugs just lower inhibitions.

Actually, there's a beta blocker called Propanolol that's often prescribed for people who have such things as fear of heights or stage fright to help overcome these things. It works by altering the way your amygdala works, that lizard part of your brain that controls fight or flight response, but it won't kill those fears completely.
