Cross tolerance between DOI and mescaline?

Just as the title says, would there be any cross tolerance experienced between DOI and mescaline or peyote?
 
Psychedelics in general are cross tolerant (whether ergoline or trpytamine or PEA), and these two are both phenethylamines, so their more similar pharmalogical profile will make them even more cross tolerant than either would be with say shrooms or something. If I'm wrong in this please correct me someone.

Yes pretty much, although my gut tells me that the farther away the compounds are related the less tolerance there should be, so after having taken mushrooms for example I do think that you have the most tolerance for mushrooms, and less tolerance (but probably still a significant amount) with a 2C-X.
But looks can be deceiving. You should not look at the structural formula (the molecules) and compare them, but instead check the pharmacological profile and see how they match up. The reason for this is that molecules that at first sight do not look alike have something in common: they fit the same way into a receptor like two keys in a lock because they have the right properties at structural positions that matter for this receptor binding.

Tolerance is tricky. I would personally take the starting assumption that there is tolerance, then I would not remedy this by taking a much higher dose unless I was ready to take that entire dose full-on... instead I would just wait out the week. Because on some occasions people do trip with only 2 or 1 days in between or consecutively. Normally the effect is really dumbed down but every now and then set and setting align in a way that for whatever reason there is little noticeable tolerance. I've read accounts from bluelighters who experienced this. The chance of this happening seems (again) bigger to me if the two psychedelics don't really act very alike.

In conclusion: peyote and mushrooms are classic psychedelics. I'm not sure if it's just the illusion the course of history would have us believe but the classic psychedelics seem to have the full package even though they are not chemically alike while many analogues are specialized parts of the package. I think this is related to high 5-HT2A affinity, looking at 25C-NBOMe which felt acid-like to me. Also Brdragonfly has high affinity and is reported to be acid-like. Classic psychedelics and/or ones with especially high affinities can be expected to be cross-tolerant due to acting alike, and also to relatively produce a lot of tolerance: look at NBOMe compounds - it is said that you need to wait two weeks in between instead of one! While something milder like 2C-B can be used multiple times in a week although it is not recommended.
The high affinity is (correlated to) stronger binding having the consequence that the receptors are more strongly downregulated. Less receptors for a drug to act on means less effect. This is how tolerance works.
