Anyone tried a tianeptine MDMA combination?

Sounds like an interesting theory to enhance the re-uptake of your serotonin, but also will probably enhance any damage that MDMA has already caused you.

One of the nice things about tianeptine in comparison to other antidepressants is that it's rapidly evacuated from the body (half life's 2.5 hours according to Wikipedia). If you want to roll, it's likely that you'll only have to discontinue use of your antidepressant for a short while (e.g. a day or two) rather than 1-3 months like you would on Prozac. Another nice thing about it is that it doesn't occupy cytochrome P450, which is used in MDMA's main metabolic pathway...a lot of other drugs do, which tends to make both drugs' pharmacokinetics a little messed up.

Standard disclaimer applies: if you're clinically depressed, you may want to avoid MDMA because it might make your condition worse. Discontinuing your antidepressant medication in order to roll may also have negative consequences.

As for what would happen if you took tianeptine and MDMA at the same time...my assumption (based on nothing but Wikipedia's pages on Tianeptine and SSREs) is that it wouldn't significantly affect your roll. Tianeptine activates glutamate receptors to indirectly cause SERT to be more efficient rather than binding to SERT directly. This means there's no competition between MDMA and tianeptine at SERT: MDMA can reverse SERT and pump serotonin into your brain as it normally does. Tianeptine works as an antidepressant by raising the relative level of dopamine compared to serotonin, so it's likely that MDMA will block its antidepressant effect by raising serotonin levels. Then again, you'll be rolling, so you probably won't notice this. *shrug*

Once again, just a theory, and one that you can review against Wikipedia with ease.

This raises another interesting question that I remember people on the forum discussing a while back: does serotonin depletion potentiate dopamine? It seems that, given tianeptine's method of action, it does. This would explain why dopaminergic activities like smoking marijuana and having sex would feel better on the comedown/the days following MDMA -- the reduced levels of serotonin allow dopamine to dominate.

Just an idea, but it would explain a lot. :)

And yeah, this may be more appropriate for ADD...
 
I suppose it depends on how much tianeptine enhances serotonin reuptake, or if it even allows reuptake to occur when MDMA is present. If tianeptine is so effective that it causes any remaining unbound SERT to evacuate the synaptic cleft faster than MDMA can fill it, then yes, I imagine it would mute your roll.

This is all speculation, of course; I guess we'd have to have someone try it to know for sure. In any case, tianeptine is cleared so quickly from your system that it's unnecessary to ever have to mix the two...it's administered in three doses throughout the day specifically because its effects are so short-lived. *shrug*
 
Allright thx for your responses, i tought it would be possible that it significantly potentiates a roll because it speeds up SERT wich MDMA binds too.
 
Instead of COMBINING it with MDMA, maybe instead use it for short durations BETWEEN rolls? Would there be a benefit in that? (recovery, tolerance reduction, enhancing the 5HT transport?)
