Could you possibly explain your analysis of the Ki values, in relation to their pharmacodynamic, pharmacokinetic, and psychoactivity?

It's cool: most of the conversation in here is speculative anyway. It's just also important that we scrutinize our speculation.

To measure binding affinity, the test ligand's activity is tested against against a radiolabeled hot ligand: the hot ligand binds tightly to the the receptor, and as the test ligand competitively binds to the receptor (increasing dissociation of the hot ligand), the radiolabeling provides an indication of how concentration of the test ligand relates to receptor occupancy. The Ki(50) value indicates the concentration of test ligand in solution necessary for 50 percent occupancy of available receptors. All else equal, compounds with higher binding affinity will be more potent, but other factors, like bioavailability, BBB penetration, efficacy at the given receptor, etc., influence potency too. However, compounds with Ki(50) greater than the micromolar range are rarely considered active (hell, micromolar range binding is still very weak).

i wouldn't know where to set foot with 6-fluoro-dmt yet inside of my mind.
I just imagine it being far reaching.
 
Todd Skinner talked the biggest load of shit. I wouldn't cite him as proof of anything.

Trust me, you don't want to trawl through the literary abomination which is Krystal Cole's book to draw the same conclusions that I did. So spare yourself some pain and take it from me.
