Hellos! SWIM has past experience using cocaine a few times, never been an often user of it because it just simply isn't their thing, but they've gotten it a few times and would use it over a day about.5 to .75g over a day period. They've done this maybe 8-10 times in their life and are interested in a clinical trial for cocaine and during the study a dosage of 20mg & 40mg would be administered intravenously, are these really high dosages of pure cocaine for someone with no current tolerance? Thanks for any info or suggestions!
 
Ritalin (Methylphenidate) is nearly identical to Cocaine in its MOA and nearly twice as potent as Cocaine at inhibiting the DAT dopamine transporter

ED-50 0.25mg of HCL salt / per Kg of body weight ....effective dose to inhibit 50% of DAT

CIBA Pharmaceuticals had a 100mg glass jar of pure Ritalin HCL. Administer 10mg IV and thats a decent dose for an adult male. Oral Ritalin is 20mg tablets X 3 daily (every 3-4 hours) ....when IVed Ritalin is MUCH more potent. As a 240lbs 6' male I usually take 60-70mg IR at once with Alcohol to feel euphoric, no more than that

A medical study with Cocaine dependant users had trouble differentiating the difference between Ritalin and Cocaine via IV injection

In a professional medical study with trained staff I'm sure it's relatively safe regarding dosing, body weight, cardiovascular health, DDI's, etc, etc

Illicit Cocaine of unknown chemical makeup or purity I'd say IV administration is very dangerous. Each batch of illicit Cocaine would have to be slowly tested each time to gauge potency and reaction to each batch

20mg of HCL salt in a 1cc 30 gauge insulin syringe and slowly press in 0.1 ml or even half that...and feel for 30 sec to a min any effects.....slowly press, each 0.1 ml until Cocaine effects are gradually felt.

Again, very dangerous to inject an unknown powder ....I'd send it to "have your drugs tested" if you live in Canada.

What's the clinical trial for? They have to be testing it for a specific reason, and cocaine can't be patented, especially in IV form, so I can't see how they could possibly get IRB/DEA permission to just hand out coke shots. I very much doubt there is a "clinical trial."

It is a trial to determine clinically meaningful interactions between mavoglurant and cocaine. Wouldn't even be a consideration if it wasn't in a medical setting. 🙂
 
Yes, Cocaine isn’t a drug prescribed for ways we might thing.

Although it IS FDA approved under the trade name Numbrino (Cocaine HCL 4% concentrate) topical solution

This was just a medical journal experiment testing Ritlain (Methylphenidate) against Cocaine HCL administered IV injections in Cocaine dependant users. Probably some kind of “Agonist Replacement Therapy” for Cocaine dependency

There's a site where you can search for specific clinical trials worldwide, I'll post it if it's not against the rules. The place I go to does a lot of recreational trials. In the past year they've done psilocybin, oxycodone, kratom and 5-meo-dmt & right now they're doing phentermine and cocaine. I was really looking forward to the 5-meo IV study, but unfortunately it was a healthy volunteer study, I didn't qualify simply because I had done recreational studies with them already. I guess in their eyes you can't do drugs and be healthy?🤷😂

I imagine if Cannabis is moved to a lower schedule they'll likely be doing more studies involving that as well.

At my old clinic they gave Vyvanse to people on meth
 
Vyvanse (Lisdexamfetamine) is the perfect drug for Agonist Replacement Therapy. It MUST be taken orally and is rate limited by enzym in blood stream to metabolize into API being active d-Amphetamine, with a 3 hour onset and 10-12h duration, slowly releasing d-Amph over the day at rate limited even flow

Vyvanse 20mg is exactly 5.9mg d-Amphetamine (as per Shire website)

Dexedrine 5mg tablets & Dexedrine Spansule 10/15mg XR capsules would be ideal
Adderall IR tablets & XR capsules even would be good

Desoxyn (d-Meth) 5mg tablets would be the best if lucky enough to be prescribed (5-25mg daily dose)
 
Depends partially on genetics, physical state, what you would expect as reached state from "high dosage", injection speed

Personal exp
Heavy tolerance
The best coke i could get, barely adultered ( free testing here )
First injection of a day after full rest

0.10-0.13g is enough to give me a crazy rush with strong bell ringer

0.04 would give me stimulating effects, but not the enjoyable euphoric rush i look for

So would 0.02 or 0.04 be high dosages using pure cocaine on somebody without tolerance ? In scientific, monitored context , maybe not that much , tho 0.04 probably already enough for some strong effects
