Design, synthesis and pharmacological evaluation of certain GABAB agonists
Abstract
Synthesis of (RS)-5-amino-3-aryl (methyl)-pentanoic acid hydrochlorides, 3 aminomethyl-5-chloro-benzoic acid hydrochloride and (RS)-4-amino-3-(4`-ethynyl(iodo)-phenyl)-butanoic acid hydrochlorides have been accomplished. The aim of their synthesis was to evaluate their GABABR agonist activity and to derive a model which will correlate their structure with the observed pEC50. The GABABR agonist activity of the prepared compounds has been determined in functional assay based on calcium measurement in vitro using tsA cells transfected with GABAB1b/GABAB2/Gαq-z5. Reviews on the neurotransmitter receptors (ligand-gated ion channel receptors and G protein-coupled receptors), their agonists and antagonists have been given in the general part of this work. A detailed discussion on the strategy followed for the synthesis of the designed compounds as well as the starting materials and intermediates has been described and illustrated in Schemes 2-6. The synthesized compounds were evaluated for their GABABR agonist activity. Furthermore, these compounds were docked in the available 3D homology model of GABABR using the program FlexiDock implemented in SYBYL software. Subsequently, we derived a predictive model which correlates the experimentally determined pEC50 with the calculated binding energy of certain baclofen analogues and homologues. In addition, we used the program DISCO (DIStance COmparisons) implemented in SYBYL software to find the pharmacophore features of GABAB agonists.

I'm still claiming beta methoxy 4-methylmethamphetamine is one day gonna be the biggest commercial hit i could imagine after comparing 2c-d with BOD. 4-MMC was so good it's now considered a 'new classic' by some and very likely to never, ever, ever stop being manufactured in illicit labs. From what i have read bk-2-cb sounded more like BOB than 2c-b (different duration and effects) so that's one reason that after sampling both 2C-d and BOD i think beta methoxy-4-methylmethamphetamine will be more like it's desmethyl, ketone relative 4-methylmethcathinone. Also, it's unlikely to produce the highly cardiotoxic metabolite 4-methylephedrine. It's also likely to have a longer half life.
Of course beta methoxy MDMA is a possibility just like methylone aka bk-MDMA was. It's likely to be similar! I prefer 4-MMC to bk-MDMA but used to mix them regularly. The
next i am interested in is BM-MDMA beta methoxy or in Shulgin's terminology BOMDMA, or since i prefer 4-mmc to MDMA, BM-4MM or BO-4MM.
I noticed @Rectify posted some beta methylthio compounds. I am truly curious about how active those would be as well. Of course I'd be interested in any possible triple releasing agents or psychedelics that could result from beta methylthio.... of course with beta methylthio straight stimulants are a possibility too.

I wish i had the technical knowledge to attempt to synth it myself. The craziest thing I've ever tried to synth myself is methcathinone. It only turned out as snortable crystal once. Every other time the reaction was too warm and got black goo. That shit has to be starting to form ice crystals when you add the KMnO4 or lots of product will be ruined.
