omeprazole and benzos

I have been taking 20mg of omeprazole for 3 days now (1 tablet per day) and I read from wikipedia about how it can potentiate diazepam through cyp3a4 inhibition.
Yesterday I took 20mg of diazepam, 1mg xanax XR, 1,5mg xanax XR crushed and 20mg temazepam. Then today I took 1mg xanax XR crushed into about 5 pieces and since grapefruitjuice potentiates xanax I was wondering if omeprazole also potetiantes xanax?
 
Indeed omeprazole does potentiate the effects of xanax and most benzos in general, though I would research into dosages. Always gotta be safe. I've also read that grapefruit juice is apparently a potentiator to. ;)

It will potentiate effects, not sure how much (it'd likely vary). Be careful.
 
that's interesting, i never realised omeprazole potentiated Benzos. How long before my valium should i pop an omeprazole to get the best out of it?

A few hours, to be sure. You want the enzyme inhibition to be well initiated.

You should be VERY careful. As far as I know, black pepper contains an MAOI. It's probably piperine, IIRC.

I think it will prolong the metabolism, resulting in a longer period of effect. Subjective strength per dose should be somewhat increased. I actually suspect, though I'm not sure, that taking it with the omeprazole will reduce the euphoria effects, however slightly.

How long does 1mg xanax XR affect you?

The half-life of alprazolam is ~11 hours. Theoretically, almost half a day into it, 0.5mg should be active in your system. However, I find that it feels like it only lasts 4-6 (if lucky), unless I take a BIG dose.a
 
Omeprazole should work for benzos that don't fall into the family of 3-hydroxy benzodiazipines.
So Alprazolam is potentiated, for sure.

But Temazepam is not potentiated by 3a4 inhibitors. Cytochrome enzyme 3a4, i'm i'm not mistaken, works to add a hydroxy group to the 3 postion - so drugs that already contain that group are unaffected by inhibition of the 3a4 enzyme, like Temazepam.

that may be a bit of a simplification though.
 
If 3A4 is being blocked, more drug molecules are forced through other CYP families cytochromes. 3A4 metabolizes diazepam to an active compound of a long half-life that is virtually as potent as diazepam itself - nordazepam. Other metabolism routes are: diazepam -> 4'-hydroxydiazepam -> diazepam-4-glucuronide or it might be hydroxylated at position 3 to give temazepam and other compounds analogically as with diazepam.

Diazepam doesn't seem to be a drug metabolism of which you don't really want to slow down.

Besides diazepam wears off quick compared relatively to its long half-life because it's a lipophilic molecules and though it can cross the blood-brain barrier, it also "goes away" from brain to cumulate in fatty tissues.
 
I have found that Modafinil 300mg seems to have raised my blood levels of diazepam(45mg a day) as well.
But in a short time, that stopped..(or I got used to this new blood level)
My Nexium doesn't seem to interfear with it.
 
If u take a few Tylenol before your nighttime sleeping aid, if its a "big one" that will increase the tiredness as well. Obviously, since Tylenol has to be metabolized in the liver.
I take Seroquel 75mg IR and 15mg Diazepam at night but if I take 1gram of Tylenol with it it feels like 20% more sedating.
 
long term use of tylenol is probably not the best suggestion (at least for those who take sleep medication every night)

The maximum dosage at any given time would be decreased from 1000 mg to 650 mg, while combinations of paracetamol and narcotic analgesics would be prohibited. Committee members were particularly concerned by the fact that the present maximum dosages of paracetamol had been shown to produce alterations in hepatic function. ... the FDA also is requiring manufacturers to update labels of all prescription combination paracetamol products to warn of the potential risk for severe liver injury.

(paracetamol = acetaminophen = generic tylenol)

so yes the tylenol may alter your liver function which has certain effects for benzo usage but I don't think that's a good thing in the long run.
I wouldn't take 1g tylenol nightly for more than a few nights in a row.

I'm now at the stage Where diaz doesnt have any effect on me whatsoever, which sucks big time!! I've tried omeprazole, grapefruit juice, even a few whiskeys when dropping them.... Nothing! Time for a break i think.
 
For the guy who doesn't feel anything anymore off of diazepam, i can relate. I have a huge tolerance but recently I have been taking my. Valium sublingualy. It kicks in faster and harder. One more note, I have done a lot of research on benzos and on the omeprazole subject, it has been stated in most monographs that omeprazole potentiate diazepam and midazolam. Or maybe it was. Triazolam but it states that only the two are studied and verified. Check a benzo in a drug interactions tool. Along side. Omep and you will see.
 
anyone know if omeprazole will potentiate klonopin???

I have started taking 40mg omeprazole a day and when taking a 2mg Rivotril (UK version of klonopin) yesterday it did seem stronger, but could of been placebo, as I don't know if clonazepam is one of the benzos that are potentiated by omeprazole.

Yes, omeprazole blocks the breakdown of clonazepam through inhibition of the enzyme that metabolizes it. It definitely potentiates clonazepam and it's not a placebo.
