1 week ago I ended up with a script of tramadol. I'm trying to take a couple days off since I have been doing them every day so my tolerance will diminish. I've noticed that after my initial dose every subsequent dose after the initial yields less and less effects. I was wondering why this occurs. Also Is there is a more proficient way to re dose?

Plugging BA isn't much higher than oral, I think it's about 70% oral and 75% plugged. Might be confusing it with another drug, but I think Tramadol's BA actually increased with repeated dosage, so theoretically, doing lots of small doses over a period of time would be most efficient.

^^^^

Like Naon mentioned, taking small doses over a period of time would increase the "high", if thats what you want to call it.

Cool. thx for the advice.

Small doses of tramadol spread out during the day are more effective than one big dose because it allows a higher percentage of the tramadol to be converted to o-desmethyltramadol, the metabolite responsible for tramadol's opioid effects.

Bioavailabilities megathread
- as nAON said rectal is not much higher than oral.

Remember not to exceed 400mgs in 24 hours.

Okay I see that you say not to go over 400mgs a day but I was just reading this other greenlighters post he says he takes 1000mgs a time and it's awesome he has been doing it a couple times a month for three years and never has a sezuir he didn't even know about the issues. Also that his friends take 350mg to 500mg and they also have had no issues.

So I guess my questions is can I take more should I take more, I wanna take more....

You be safer just using a better Opoid, im not saying that the other Opoids dont carry a whole lot of risks but to my knowledge Tramadol is the one thats known for seizure.

People differ with their seizure threshold. We had a poster here who seized on less than 400mg. Just because one person is okay with higher amounts doesn't mean you will be too, and seizures can be fatal.. make sure you avoid any other medications which can also lower the seizure thresold and you really should not exceed 400mg/24 hrs.

The answer to your question is that no you shouldn't take more - it's impossible to know if it is safe or not, but the consequences of it not being safe are pretty severe.

100mg attack dose, with 25mg boosters every 20-30 minutes until a total dosage of 250-300mg tramadol is achieved. This method of staggering your doses ensures that the maximum amount of O-Desmethyltramdol is converted and cuts down on the seizure risk by preventing a spike of stimulant activity.

Going over 400mg a day is incredibly stupid, as people have been reported to seize at doses of 250mg.

^ I believe it is because it gives your liver more of a chance to metabolise tramadol into o-desmethyltramadol, which is responsible for tramadol's opioid activity :)

I used to get 100's of those. I'd take 8, wait an hour and take 2-3 more. Proceeded to do that until I felt nice. I used to end up doing 20+ in a day but that's because I had a high tolerance.

^ you need to be careful with tramadol - it is not recommended that you take more than 400mg/day, even with a tolerance, as it has SNRI action as well as opioid action and there is a very real risk of seizures. People's seizure threshold varies, but we had a poster a while back who had seized off less than 400mg and just because you've been okay in the past doesn't necessarily mean you will be in the future so do be careful..

^ I would imagine so yes, the enzyme responsible may become saturated. I would need to read up on it as well :)

PSA: Always do a 100-125mg dose of tramadol if you've never done it before, as you might be CYP2D6 polymorphic and not know it, as I was. 125mg blew me the hell away; if I had treated it like the weak opioid that most people say it is and did 300-400mg, I might have overdosed.

Most people aren't 2D6 polymorphic (have multiple copies of that enzyme), but don't assume you are like most other people. 2D6 polymorphics convert a much larger amount of tramadol in to O-Desmethyltramadol than others with normal or deficient 2D6 enzymes.

By that logic, the more CYP2D6 deficient you are, the more gradually you should redose. For the heavily 2D6 deficient, maybe 25mg every 45 minutes instead of 20-30 would be ideal?
