I didn't see anyone correct this yet, but, The Big Lebowski last line is:

The last lines in the movie "Stand By Me".

Made me tear up the first time I saw the film.
 
There are more lines before leading up to this quote but this is the one that puts the lump in my throat. "I never had any friends later on like the ones I had when I was twelve. Jesus, does anyone?"
