What is the mechanism behind burnout?

Some marijuana users seem to "burn out" after prolonged heavy use. I don't know a more scientific way to put it but you guys know what I mean. Does anyone know what is the mechanism behind this?

Actually I really can't tell what you mean by "burn out" based on your post.

Do you mean they don't get the same kind of effects from using as when they started?

or that they lose motivation and no longer achieve on the level they're capable of? or something else?

In any case I doubt anyone can put a precise neurochemical mechanism on such a high level process like "burning out".
 
Thc is a partial agonist of 5ht2a, it probably puts stresses on the 5ht2a mechanism that regulates neurogenesis.

That is my somewhat uneducated hypothesis
 
Is THC? I've never heard this, and the closest I could come was this article, which only suggests but does not directly demonstrate that some effects of THC might be mediated via (likely downstream) activity at 5ht2a (Egashira et al. 2007). Also, THC itself promotes neurogenesis localized in the hippocampus.

Well, wikipedia has references, which you can look up to verify and elaborate on entries.

edit: found the article: "Agonistic Properties of Cannabidiol at 5-HT1a Receptors" (Russo et al. 2005).

They found CBD to act as a modest partial agonist at 5ht1a (micromolar range affinities, similar to its affinity for CB1) and bind to 5ht2a, but far more weakly, with unassessed effects on signal transduction. THC itself, though, does not bind to 5ht1a or 2a sufficiently tightly to be relevant. Oddly, anandamide has also been shown to act similarly to CBD at 5ht1a and 2a.
