I'm currently on 75mg of Effexor (Venlafaxine) a day, along with 40mg Vyvanse. I've recently come upon some Tramadol 50mg pills. Is there a great risk of serotonin syndrome if I take 1x 50mg Tramadol?

Vyvanse (a pro-drug for dexamphetamine) only has minimal serotonin action, but vanlafaxine is another story.

Mixing two serotogenic drugs together is generally not a good idea and can be considered dangerous.

I've been on Vanlafaxine for some time, 150mgs XR daily, you really should avoid Tramadol I'm sure NT is correct but Venlafaxine also lowers your seizure threshold. As you may know Tramadol can bring about seizure at doses higher than 400mgs in 24hrs, in some at even lower doses so the 2 together are not a good mix.

I've recently sustained a nasty knee injury and can't take them at all so am taking MS Contin instead.

Even at these low dosages for both drugs, there is still a high risk of Serotonin Syndrome?

I'd be much More worried about the seizure risk than seretonin syndrome. Vyvanse and tramadol is a bad idea and could be catastrophic. Codeine would be a MUCH safer choice of pain relief.

^ Agreed, amphetamines can lower your seizure threshold, as can venlafaxine, and tramadol has a well-documented seizure risk. I would not combine the 3, even at a low dose of tramadol - people vary widely in their response and it's not worth the risk (especially considering the minimal benefit you would probably get from 50mg tramadol).
