I've read here that Tramadol would be TAAR1 agonist. However, I didn't find any info or studies concerning that matter.

Has anyone any studies on TAAR1 and Tramadol? Or any other information regarding that function?

Any information is appreciated and needed.

Thank you!

There is no good SAR based reason to think tramadol is an agonist at TAAR1 nor is there any evidence in the literature to suggest it interacts with TAAR1 in a clinically relevant manner.

Any substance that is similar to Tramadol in effects but causes less sedation ? I was doing fine with 200 mg of Tramadol daily (apart from sedation), now I am on a break and considering my options for the future. SSRI are not so great for me. I would avoid havyweight opiates.
Thanks

If tramadol causes sedation for you then you will hate the other opiates since tramadol is the most stimulating one due to SNRI action. Id say your option is to find a similar SNRI med since it looks like you're not using tramadol for the opioid effects

I suggest that you try tapentadol to see if you like it. It is class A though whereas tramadol is only class c.

You could be right. I guess I enjoy both. But I definitelly don't like to be slowed down with Tramadol, and it happens.

Venlafaxine is an SNRI that works similar to tramadol but obviously without opioid effect (which is maybe the sedation you're talking about ). With venlafaxine you will be stimulated with 0 sedation. I was prescribed that shit before but I hate ALL anti depresants soo I prefer tramadol for it's opioid effects but it's nice to be a bit stimulated aswell

Or if you're into opiates there is some with stimulating properties like oxy. Any opioid synthesized from thebaine will have stimulant properties on top of their opioid action but like you said you don't like sedation and it might not be for you

Where did you read that Tramadol might be a TAAR1 agonist, if I may ask?
