The natural compound Borneol found in Nutmeg is a potent positive modulator of GABAA receptors, and studies have shown its modulatory effects can be more efficacious than some benzodiazepines. It is also one of the many components found in nutmeg essential oil.
Borneol and GABAA Receptors
Presence in Nutmeg
Borneol is a bicyclic monoterpene found in the essential oils of many medicinal herbs, including valerian and, as you noted, nutmeg ([Myristica fragrans Houtt.]). The essential oil of nutmeg contains borneol, along with other major compounds like myristicin, sabinene, and alpha-pinene.
I got this from Google by the way.
Borneol and GABAA Receptors
- Mechanism of Action: Borneol enhances the action of GABA, the primary inhibitory neurotransmitter in the brain, at the GABAA receptor. This leads to increased chloride ion influx into neurons, making them less excitable and producing calming, sedative, anxiolytic, and anticonvulsant effects.
- Potency vs. Benzodiazepines: While its overall potency (effective concentration) may be lower, the efficacy of borneol in modulating GABA responses (the maximal effect it can produce) has been found to be equivalent to potent anesthetics like etomidate and greater than that of diazepam (Valium®) in in vitro studies using human recombinant receptors.
- Different Binding Site: Unlike benzodiazepines, which bind to a specific modulatory site, borneol acts at a different, distinct site on the GABAA receptor, as its effects are not blocked by the benzodiazepine antagonist flumazenil.
