Studying structural activity relationships

I was wondering if anyone hear had any good starting points, literature, research articles, etc. that can help me learn more about the role structure plays in a psychoactive's pharmacological properties?
You all seem to be well-researched and can make speculations about activity and toxicity of even completely novel compounds, where did you start off?
I'm already versed in introductory organic chemistry and biochemistry directed towards enzyme catalysis, also slightly in neuroscience, I have also read tons of papers on pharmacological properties of various drugs but they're often so in depth it's hard to pull out the more general concepts embedded within it.
Any guidance would be appreciated :)
 
Learn organic chemistry, then learn a bit of biology and medicinal chemistry. Then just look at a set of similar compounds and you'll be able to piece together a lot of the SAR.
