Regular RIMA antidepressants (moclobemide) regemen and psilocybin

There has been a glut of posts about the potentiating effects of taking an MAOi substance preceding a trip, one such substance which is getting a lot of noise is moclobeminde (aurorix, mannerix, amira) due to it being a reversible MAOia and the ease of its acquisition.

However I am yet to see a thread about psychedelic substances, specifically psilocybin and habitual moclobeminde use.

After 30 days of administration, moclobemide and brofaromine selectively inhibited brain MAO-A activity and phenelzine inhibited MAO-A and -B to equal extents. All three drugs caused a significant down-regulation of tryptamine receptors, whereas only phenelzine significantly down-regulated 5-hydroxytryptamine(2) receptors.

This study seems to suggest a significant down regulation of the tryptamine receptors after prolonged use (one month +). Given the excess amount produced due to the lack of monamines in the system, homeostasis is to be expected.

Given that the down regulation of the tryptamine receptors is an indirect result of the drug, abstaining for a day or two probably would not give sufficient time for the receptors to up regulate, even if there were no moclobemide in my system (I've been consistently taking 600mg a day for 18 months).


moclobemide and LSD

According to this blog in order to achieve the full effects of LSD, which is a related chemical, he has to abstain for three weeks.


I suppose I could do an experiment, but since I only have access to enough active mushrooms for one decent trip I would rather not waste it.

Anyone have any first hand experience, or anecdotes with taking mushrooms, or any tryptamine drugs, while on a moclobemide regimen or any other MAOi antidepressants?
 
MAO-A is largely responsible for the metabolism of tryptamine drugs like psilocin(mushrooms) and DMT. Even though the chronic increase in serotonin forces a downregulation of 5-ht receptors. The inhibited metabolism of the drugs in question could make up for that loss. Most people don't use pharmaceutical maoi's for mixing with psychedelics. Plant extracted beta-carbolines such as harmine and harmaline are much more common RIMA's for potentiating tryptamines.

I'm not sure how MAO-A affects lsd metabolism though.

You might also want to look at information about downregulation with ssri's. The don't have the same pharmacology, but the downregulation should be similar.
 
So if I decide to go ahead and continue my regimen as per usual on a day i decide to trip, the MAOI effects may compensate against the down regulation, whereas if i were to abstain for a few days, I would suffer the down regulation still without the MAOI compensation?
 
The half life of moclobemide is only 1 to 2 hours, so the effects of the drug should cease fairly shortly after you stop taking it. But if taken at the same time as the mushrooms, it will potentiate it. How much is completely conjecture. Some people report maoi + mushrooms to be extremely intense. In your case, downregulation is almost impossible to measure so I can't really give you good advice. It might be a dud, or it might knock your socks off. Wait for info from someone who has done this.
 
I read that moclobemide's MAOI activity lasts 24 hours. Unfortunately I lost the piece of literature in a hard drive crash.
