Hi, I am planning to take mdma + a mild variant of magic mushrooms called 'truffles' (atlantis) In 2 days. if I usually need around 100mg MDMA for a nice roll, and 4 gram truffles for a nice truffle roll, how much should I take to combine? (I read they are synergetic)

No set time - you need to experiment to find out whats best for you. I wouldn't take the MDMA too early because once that wears off it pretty much kills the trip too so you want to take it later rather than early.
I don't think the truffles are "mild". They'll all be pretty much the same.

Maybe take 3/4 of your normal doses IMO.
I agree with Ismene: truffles are probably considered mild because the quantity in a 'standard' dose of truffles may amount to a lower dose of alkaloids. Sclerotia are about 2 times as potent, so if you normally see 10 grams of truffles in a package that would be about 20 grams fresh mushrooms (or 2 grams dry).
The rest is probably just expectations and placebo / bias causing subjective differences - as long as the quantities of psilocin and psilocybin are actually the same.
Just start with a dose of mushrooms or truffles to give a moderate but not overly strong trip, and when the peak levels off drop the MDMA.. Again I would drop at least a moderate dose. There is synergy to account for but I would make sure you don't dilute it too much by lowballing it extremely.
