nicotinic acetylcholine receptor antagonists and nicotine cessation

Has anyone had any experience with this? Or know anything. I've been trying to quite smoking and have noticed some quite interesting phenomenon. I was taking an NMDA antagonist (DXM) along with Piracetam (high dose). Stopped the DXM today and am going to continue on with the piracetam.

Furthermore, is there any reason to think that by replacing nicotine with choline it might be easier to stop smoking? If someone could explain this I would appreciate it. Thanks!

A partial agonist would help more IMO but, hey, some have quit w/ antagonists on this receptor. I gave my girl wellbutrin when she was a regular smoker, it seemed to be cig. smoker's naloxone to her and she went through precipitated nicotine WD's (if there ever was such a thing) for like half a day. She said no other pill made her so suicidal.
 
A nicotinic ACh antagonist will cause up-regulation but it may depend on the antagonist'c functional selectivity. It will induce withdrawls especially if it is not not competitive. A partial agonist with less ligand efficacy or a competitive antagonist would be more appropriate like Captain mentioned. A non- or uncompetitive antagonist would be miserable what was said and would only make sense if you had overdosed on nicotine...which doesn't happen too often.


But Piracetam is a muscarinic positive allosteric modulator so it's not going to help anyone quit smoking. The DXM on the other hand to some extent helped up-regulate those N ACh receptor sites. But how effective DXM is of doing that is probably not much and somewhat unknown.

"centrally" speaking... bupropion, mecamylamine, and 18-methoxycoronaridine block nicotinic acetylcholine receptors in the brain and can be used for treating certain addictions. other types of nicotinic antagonists are used during surgery because of the peripheral muscle paralysis effect that can be achieved.

i don't think this is really a good way to treat a smoking habit ( i assume that's what you do, smoke ). i find it best to buy a pack of harsh cigarettes like some american spirits or lucky strike unfiltered. i can barely make it through a pack. then you just have to make it through the first few days, after that it's not so bad.

addictions are a mental thing mate, not something you can just take a pill for and call it good. it's about changing behaviors and lifestyle... doing something else with your time. smoking addictions are best to fight with a partner i think. it's hard to do it myself, but if my girlfriend (or friend) and i do it together it seems easier. just fuck when the smoking urge comes on, or SOMETHING. anything but smoking! after the first few days it's a lot easier bro...

oh yes, and the best thing for reducing caffeine tolerance is simply taking a day or two off. i sincerely doubt DXM or piracetem is going to reduce caffeine tolerance.

Piracetam increases neural excitability via AMPA positive modulator so it may mediate the effects of caffeine more potent.
