Methylphenidate blocks neurotoxic properties of methamphetamine.

no i think it just means they won't suffer if they decide to stop using meth. i've never really understood this study because it seems to contradict itself. i think I've posted on this one before.

considering their mechanisms of action so:

methylphenidate NDRI

methamphetamine D+++S++N+/RI to some extent

surely would lead to larger dopamine build up in the synapses, causing more damage, frankly can't see any neuroprotection going on whatsoever here. while it MAY prevent the release, the dopamine is still being produced at enormous amounts. serotonin may have a role in this though not sure entirely.

can also see a bunch of cardiovascular implications too. like its just not worth it at all.
 
is this not the same situation with mdma (serotonin releaser) and ssris? ssri pretreatment diminishes mdma's binding at sert et cetera

if you block the ND transporters with some other ligand so that meth cannot bind/enter the cell then yes you would expect diminished damage but also diminished effects from the combination

but in reality you're not going to take a dose of mph big enough to block all the transporters when people often dose upwards of 100-200mg meth in a binge

IDK, one never can tell with a speed freak.

But couldn't methylphenidate actually potentiate meth? As I understand meth also releases dopamine via TAAR1 and the dopamine transporter, and methylphenidate will prevent this dopamine from being reuptaked
 
it's dose dependent... if you take 500mg of meth and 10 of methylphenidate don't expect any protection at all.

it's nothing but a fun mental excercise really this is not something I would advise as a real treatment.

Could one speculate that 50mg of methylphenidate and 50mg of meth could feel equivalent to maybe ~150mg of meth?
 
No: one would expect highly idiosyncratic potentiation (or attenuation of effects, in some cases), varying greatly with both sum dose of whichever given compound, the ratio of the compounds, and how dosing is timed. You could easily end up with synergistically increased cardiovascular activity (and thus strain), which is bad.
