Mannitol to potentiate drugs

Mannitol can also be used to open the blood-brain barrier by temporarily shrinking the tightly coupled endothelial cells that make up the barrier. This makes mannitol indispensable for delivering various drugs directly to the brain

Has anyone got any experience with this?
 
Id really avoid fucking with the BBB. It's there for a reason and having it removed allows all sorts of crap into the brain.

Never less im intrested in other peoples opinions.
 
In theory this is possible, although I know solely of trials with animals (mainly rats). Fucking up your BBB is highly discouraged!!!

Interesting, I would like to read more. The wikipedia page has no reference for the fact that mannitol is used to bypass BBB.
 
For experiments with rats, a usual protocol employs direct injection of 25 % mannitol solution into the carotid artery. This reversibly disrupts the BBB to such an extend that even certain enzymes (!!!) are able to enter the brain. I can't comment on side-effects of this procedure, as mostly the rats are killed shortly afterwards to study the brain or whatever organ is of interest... :\

Everyone who intends to inject mannitol - doesn't matter into which vessel and and at what for a concentration - hits my personal definition of a 'suicidal idiot'. Just to make that clear. As far as we talking about theory, I'm fine with this topic.

^...errrm and it doesn't trouble you that these kind of treatment is described solely (!) for patients with serious forms of cancer? I strongly doubt that it is not dangerous!
 
This sounds even more reckless than some of the page-long drug regimens I've put together.
 
Shouldn't we be more cautious of such supplement if its taken via intravenous route?
The bioavailibility is only 7% otherwise


Though I heard about "blood brain barrier weakening" agent with other drugs too, Whether it was DMAE or DHEA, i forgot but then i read further that they used it with 5-HTP and other large particle drugs such a GABA which does not seem to cross the BBB that easy as its fairly large particle size.


Anyway, i went along and typed "Mannitol and Dangerous" in to google search engine because if this is so "Dangerous" then why cant it be bought over the counter.... I thought to my self

CONCLUSIONS: Long-term administration of mannitol can induce significant increases in cerebrospinal fluid osmolarity in patients with subarachnoid hemorrhage or severe head injury. This may be an undesirable and potentially dangerous effect. Therefore, cerebrospinal fluid osmolarity should be measured regularly in all patients receiving mannitol for longer than24 hrs. If cerebrospinal fluid osmolarity increases, discontinuation or tapering of mannitol therapy should be considered.

Though MurphyClox seems right as its a hard drug used for people who are in serious condition already...
But probably in other routes other than oral.

As i read on about it being poorly absorbed by the body hence why it is used as a.. laxative in high dosage, sweetening agent for diabetics who do not favour a spike in insulin levels, and also used in most chewing gum, along with sorbitol and xylitol

Mannitol hexaacetate is found in heroin when mannitol is added to the morphine prior to the acetylation step.
Mannitol is also found as a cut in heroin, cocaine, and methamphetamine. I can't imagine that injecting the amounts found in recreational drugs is going to have this BBB permeability effect.

5HTP and GABA do not cross the BBB mainly due to their polar character, not due to their size. Non-polar molecules of this size easily cross cell membranes. You were misinformed buddy!

It probably won't, your imagination is correct. The needed amount would demand intravenous or intraarterial injection of relatively high concentrated infusions, and that is usually out of question.
