How good is hydroxytyrosol for MAOI-B?

Hydroxytyrosol is reported to be a "potent" natural MAO-B inhibitor but it's plasma 1/2life is under 3hrs and it's ic50 for MAO-B is (16.3 + 1.3 μM)-i have no clue what that equation means, i'm assuming the nanoM means it's within a reasonable dosage range?

So if someone wanted to ingest it for MAO-b inhibition suitable enough to make a non orally active phenethylamine active like phenethylamine hcl, how much would someone need to ingest of it and how long would it work for(on average)

Also would there be a way to extend it's life by inhibiting a CYP450 enzyme, and if so, which ones?



***:tea catechins are similar in structure but are far weaker although more renown for MAO(B)I

Anything would be greatly appreciated.
 
micro-molar MAO inhibition does not bode well for pharmacological actrivity - it's 100 to 1000x too weak

nanomolar MAOB inhibition is seen with segelegine, look how potent that is. Many of the flavonoids like rosiridin are also micromolar MAO inhibitors - they don't do a good job. Even piperine is a maoi, just not a very strong one
 
well hydroxytyrosol is actually in the nanomolar range if i'm not mistaken, but I don't know why it's represented as an equation in this case rather than just 1 solid number?
 
It's in the micromolar range because μ means micro, not nano

The number is represented as a (mean +/- standard deviation), like many binding assays. It's a way of indicating how big the error is likely to be in the measurement of Ki. So hydroxytyrosol really has a Ki somewhere between 15.0 and 17.6 uM.
 
Half life doesn't matter. Once it's bound to MAO it's no longer seen, making it look like it's no longer present.
 
Given the comparable affinity =of e.g. piperine, I would wager a "dose" of this is more than 50 grams of ingested material
 
wow, well thanks. I don't understand why ppl in the research world throw around words like "potent" then when this is anything but, just goes to show how a degree and peer reviews don't necesarily equal intelligence IMO
