to make 4-Hydroxy-5-methoxydimethyltryptamine
i was wondering how much amt i would add to the substrate

Well some references would help, something along the lines of "adding tryptamine to mushrooms increases synthesis of X".
I'm not sure where this is as far as synthesis discussion. But, I'm not convinced it would work anyways, biology is kind of picky on substrate.

As mushrooms natively produce tryptamine, one would need to supplement with e.g. 5-MeO-DMT or the like to get 4-HO-5-MeO-DMT. AMT or plain tryptamine will not work.
I thought Shulgin once mentioned it would propably be possible to add X to the mushroom substrate, and then the mushroom would 4-hydroxylate that.
Adding MiPT would produce 4HO-MiPT, adding DET would produce 4HO-DET, etc.
So, in theory, aMT would yield 4HO-aMT ?

Goddamn I'm getting out of the loop (old? Hardly!) And Gartz is still going strong. I was going to comment on how hilarious it is that ancient conjecture by Shulgin has turned into gospel in the ensuing decades (!) but lo and behold Gartz is still kicking ass. . . Now if he would just slip John Allen some "new non hygrophanous non bluing with rust colored spores psilocybes" (galerinas) to bioassay I'd send him a trophy. . .
Gartz is a nice guy. But nice enough to rid the world of the jackass on his coattails? 
granted, Allen has done some good work for a laymen. But we come from the same block. And in the earleir days of the pioneering work on Northwest Psilocybes I had too many stomach turning encounters with him, and it appears his presence online only got worse. He's been quiet for a spell, or I've got my blinders on. Or maybe he went to the great galerina in the sky.. .
