Opioids Cross tolerance between endogenous and exogenous opioids

I've always noticed that I had an abnormal insensitivity to recreational opioids. I have also known for a while that there may be a cross-tolerance of sorts between endogenous opioids and those administered exogenously.

Today I was casually sifting through the literature and noticed (yet another) abstract that supports my above speculations:

Recent work has shown that chronic exercise is associated with a reduction in the pain-relieving actions of opioid drugs in experimental animals. To determine whether this reduction represents an interaction between exogenously administered opioids and the endogenous opioid system, or is the result of altered drug pharmacokinetics, the antinociceptive actions of morphine and its metabolite, morphine-6-glucuronide (M6G), were compared in active and inactive female Long-Evans rats. Active animals were housed in running wheels and inactive animals in standard laboratory cages for 3 weeks preceding determinations of antinociception using the tail-flick test. At the end of the 3-week period, active rats were running the equivalent of 9-11 km a day. Antinociceptive responses, determined following subcutaneous injections of either morphine (0.625-20 mg/kg) or M6G (0.3-10.0 mg/kg), were significantly reduced in active rats relative to inactive rats. This reduction was manifested by both a lower magnitude of antinociception, and a shorter duration of antinociception after drug administration in active compared to inactive rats. This reduction was not associated with alterations in the estrous cycle or with differences in body weight between the active and inactive animals. The present results support the hypothesis that cross-tolerance develops between endogenous opioid peptides released in response to exercise and exogenously administered opioid drugs.

I have always enjoyed running and especially enjoyed the incredible "runner's high" that is almost certainly caused by central mu opioid receptor activation. But whenever I use, say, morphine, I always seem to get a minimal high at best.

Now this study has its limitations in that it was performed on rats but it is well established that the similarities between rats/humans are quite profound.


So to get to the point: Are there any running-enthusiasts here that have noticed such a phenomenon when they use opiates?

Note that I am strictly referring to long-distance running since anaerobic exercise/other lighter physical activities have minimal mu opioid receptor activation as a result of the physical exertion of the activity. But anyone may respond of course!

Right on! I considered starting a thread on a topic like this, but never actually did so.

I used to be an avid runner when I was younger, before my knees gave out on me. I ran anywhere from three to six miles, three or four times a week. I'd even do eight mile loops every once in a while, just to prove to myself that I could. I know the term, or concept of a "runner's high" is a cliched one, but it is absolutely true nonetheless. I started running at the age of sixteen, when I got sent off to therapeutic boarding school, and running was the only 'high' available to me for that year.

I wouldn't say I can relate to a(n) insensitivity to exogenous Opioids, but I can say that no amount of any full agonist I take provides the sensation I get (or used to get) from long distance running. Therefore, I feel that no exogenous Opioid is capable of providing me that same feeling I got from my endogenous Opioids when I was running regularly.

No other form of exercise or physical activity, IME, provokes the release of endorphins (endogenous Opioids, among other things) the way running does, or did for me at least. If it weren't for my knees, I'd still be doing it. Running is, without a doubt, addictive in its own right.
 
Hey, thanks for the response!

It would be immensely useful if I could, say, see what 20mg oxycodone would do for me before I started running and then again two years later. That is incredibly unscientific, though.
 
No problem! Like I said, great topic. It appears as though you and I might be the only runners/ex-runners around at the moment?

As for your experiment, unscientific?;) Perhaps. But it would be interesting nonetheless.


Another thing I have noticed is how the effects of Opiates become more pronounced when taken right after, or shortly after strenuous exercise, or even not-that-strenuous exercise. For example, even if I just go on a couple mile walk before taking an Opiate, the effects are all around better in general, as opposed to taking the same dose of the same Opiate when I've just been sitting around (posting on BL=D) or doing whatever. I suspect this is directly correlated with endogenous production synergizing with the exogenous drug. And without a doubt, the more strenuous the exercise, the more pronoucned the effects of the Opioid become, IME.
 
That's actually quite fascinating as it reminds me of my alcohol + running combinations (yes, I'm aware alcohol isn't the best post-running recovery beverage).

I did this intentionally hoping for a synergistic effect and that is indeed what I received. And although alcohol isn't an 'opiate' in the traditional sense, mu-receptor knockout mice find alcohol uninteresting suggesting that there is an important link between the opioidergic system and what makes alcohol 'fun.'
 
I played soccer for 15 years (which in a sense IS long distance running if you play midfield or stopper - my position) and then switched to running. It's true, no doubt, that running creates a high unlike (seemingly any other form of exercise. It's addicting, and it's hilarious to try to explain that to 'non-runners'. I always find myself unable to STOP running when in the zone - I HAVE to keep pushing and it's not because I need to but because my body feels so damn good.

I also, in the last 4 years'ish), am an opiate addict. I can't say that the feelings associated with them are close and I wouldn't know what to say about association between the activities and mu receptors. I DO know that when running the high is there, it's real. I too wish I could go back and feel 'that high' with and without opiates for comparison, etc.

Great post for sure. I kind of want to run now (however, I too have knee problems).
 
^You and I are in the same boat, woamotive. I played soccer for about ten years growing up; I played mid-field as well. You are correct, ninety minutes of being a stopper is indeed long distance running! I know that playing soccer before I started running helped to put my knees in the condition their in now, unfortunately.

Indeed, nothing like that runner's high. And it's true, non-runners just don't seem to understand.
 
Looks like I'm late to this party. I know that the runner's high is real. Surprisingly, sometimes even other runners don't understand this (not all runners get it). I never did (and still don't) do illegal narcotics; the running itself produced the real high. After about 15K, I would often get a feeling of bliss and that all was well with the world.

But these sensations don't seem to last for long. I haven't gotten the mental benefits from running for some time. 900mg St. John's Wort, perhaps alone or perhaps interacting with a low dose of a RX antidepressant, may help retrigger these feelings during and after a run. They did for me, at least, for about 8 weeks. But it doesn't seem to last...
 
hm..... what about dancing?
i have horrid knees and stopped running back in high school..... but dance has always given me that same "endorphine rush" as running, perhaps because it is cardio in its own right, and i dance(d) for 4-5 hrs straight 5 days a week plus some light dancing on the weekend.
and i have to say i did notice a decline in effectiveness of opiates.
i also ride competitively (hunter/jumper), which is a GREAT high.... but i haven't put this to the test against opiates. hm.

similar experiences?
