Codeine and amitriptyline metabolism

I know that both codeine and amitriptyline are metabolized by the cyp2d6 enzyme, so i was wondering do either of these drugs affect the others metabolism?

I asked a pharmacist about any possible interactions and he just said the usual about increased drowsiness and such. When i asked him if either of the drugs had an affect on the way the other one was metabolized he looked at me as if i had 2 heads and mumbled something like i dont think so.

Any info from you guys would be appreciated. Thanks in advance.
 
Pharmacists... geeze... Codeine only messes around with CYP2D6 when you're up around the 60-100mg mark... and amitryptaline is relatively dependent on CYP2D6 [1, 2, 3] Amitrip inhibits CYP2D6 [4, 5], though to what extent, I can't find out. Though I doubt it is very much.

So in conclusion, they both inhibit CYP2D6, but codeine only at high doses, and amitriptyline, probably not very much. I wouldn't be concerned.
 
Thanks for the info. I always wondered if there was any interaction besides the usual increased drowsiness and i dont always trust those pharmacists. I didnt want to be decreasing the effectiveness of either drug.
