So all 4 enantiomers can be resolved (or a chiral precursor used). I would be looking at patents behind the people who own ZINC. If I had to guess, I would suggest ZINC1670065 might well be as active as dexamphetamine.

Personal experience (when I had delusions of being a competent synthetic orgnic chemist) was that replacing the beta methyl group with a beta methoxy, gives a bloody good stimulant and the configuration about the beta methyl carbon doesn't have that much influence on activity (aka O-methylephedrine. Think of it as halfway between meth and phenmetrazine - it's like a ring opened phenmetrazine).
 
THANK YOU. Those ARE novel so I could not find them. But you cannot argue with personal experience. So will we see people trying to work out how to selectively O-methylate ephedrine?

Actually, I'm sure we both know EXACTLY how to make the class you describe. Of course, after you with the alkylating agents... I insist ;-)
 
Well strangely - if it's a patent then it's in the public domain anyway. Let's face it, how many people would actually pick through a patent? Over the years they keep changing terminology and so it's a very long-winded business. How many people on BL would spend a whole day reading and understanding a patent?
