Opioids At what point should you take Benadryl if youre sensitive to histamine in codeine?

Basically should you take the Benadryl before, with, or after taking the codeine? I hear the Benadryl can dampen it a bit but at the same time I get a pretty massive histamine reaction at even 120mg, previously I took the Benadryl a little too late I think. Advice? Thanks.

It is said antihistamines affect the CYP2D6 enzyme which is resonsible for the unwanted metabolites of codeine, such as nicodine, etc. In my opinion if you want to make sure that CYP2D6 metabolism doesn't lessen the codeine, I would take the benadryl or whatever antihistamine after I know the codeine has fully kicked in, so that it will give the extra sedation without taking from the codeine-morphine conversion metabolism and the same for benzodiazepines as they too involve the same enzyme, though because of their sedation you would never gess they 'lessen' the actual effects of codeine (coverstion rate, unewanted metabolites).

I have tried to potentie codeine in an effort to replicate Doriden (which was banned before I could try it) I had phenobarbital and amobarbital in doses as high as 100mg of each to induce CYP2D6 as people are of a school of thought that makes them think inducing the CYP2D6 but got none of the pomised potentiated effect if anything a lessening of effects. Doriden worked a lot more different than just the CYP2D6 metabolism. I have found Soma to be a very good potentiator, better than benzos yet very short lived, very short, re-dosing every hour short!

Short answer for the O.P. take the benadryl/antihistamine when you feel the codeine has already taken effect, for me usually after 45min-60min.

BTW on a side note I was under the impression that promethazine was/is over the counter?! Esp in Canada yet was told I need a prescription, isn't there an OTC syrup with promethazine/codeine in it? I know this has been discussed before but are there any antihistamines on the market more sedating than benadryl? I have had an atropine containing medication (it had amobarbital in it so it was worth it) and it is said atopine has very little antihistamine/chlorinergic effect (hallucinogen inducing) as opposed to gravol, benadryl, etc and a higher level of sedation as well.
