Hi all,
From the literature, it seems that opioid tolerance is caused by the downregulation (endocytosis) of mu-receptors which is caused / initiated by the recruitment of beta-arrestin (a protein which causes the mu-receptor to recede into the dendrite)
Are there any known arrestin inhibitors, or anything which prevents downregulation of the mu-receptor in lieu of chronic activation by mu-agonists (such as opioids?)
Thanks
