I am writing to get your take on the relative safety of combining Pregabalin and MDAI. It is a very interesting combination and I cant seem to find any data or anectotal reports.

According to wikipedia, approximately 98% of RA labeled pregabalin is recovered in the urine. If I am not misstaken this indicates that there are few metabolites that causes interference. This leads me to believe that the risks of potentialy dangerous synergy with MDAI is low.

I am not well educated in these matters. IMHO it is a beautifully synergetic combo and I would very much appreciate any educated input on the potential dangers of the combination.

Much appreciated,

Anybody care to extrapolate based on known pharmacodynamics? α2δ / SSRA?

Sure,

The SSRA mechanism is straightforward, VMAT1 and SERT are tampered with, 5-HT can diffuse into the synapse.

The α2δ subunit of the voltage-dependent calcium channel is messed with by Pregabalin. The question is, is calcium diffusion through this channel enhanced or inhibited by Pregabalin? Since it seems to have anti-pain and depressant qualities, it probably decreases Ca2+ influx to the postsynaptic cell when it's excited, so the the cell gets less of a boost to an action potential.

I do not know of any interactions, and there aren't any obvious interactions as far as I know.

The only real thing i've ever worried about with pregablin and even gabapentin is that funny little neuroplasticity decreasing effect gabapentin and pregablin will have. More or less will just put neuroplastical modelling processes on hold from what I understand.

Not a problem most of the time, but it is a little strange to take NGF(nerve growth factor) promoting chemicals with chemicals that inhibit brain change...I would imagine any positive experiences taken from the MDAI would be harder to integrate ,as chemicals promoting brain growth are inhibited with pregabalin. weirdness!

On the flipside, theres probally a good amount of neuroprotection going on which is nifty when taking depleting drugs such as MDAI.
