Questions about amphetamine tolerance and neurotoxicity

Just had a few questions about these two things I couldn't fine searching the web. What does it exactly mean when you build a tolerance to amphetamines? Does tolerance make it to where the amps realease less dopamine/norepinephrine each time or does it release the same amount but just doesn't feel like it is as much with less tolerance?

And my question on neurotoxicity: Are amphetamines (not methamphetamine) more neurotoxic when one large dose is taken at once then if you were to spread it out? Say someone takes like 60mgs of adderall (not sure if that's nuerotoxic but lets pretend it is) at once. Would it be less neurotoxic to take 30mgs than 30 more a few hours later or would it still be as neurotoxic as taking 60mg at once?

For amphetamine tolerance its just like any other toleance, your body gets use to it and you need more amp to achieve the same high.
and for your second question, it would be more neurotoxic to consume 60mg at once aposed to 30mg now , 30mg later.
when taken at diffrent times its spread out and thre amp salts dont hit the brain and disolve all at once.
 
I suspect that the same amount of the chemical will consistently release the same amount of Dopa/Nor each time, but the damage to / downregulation of receptor sites causes it to feel as though the dose were reduced (lessened effect).

In terms of timing / doses - high doses are just as neurotoxic as a singular event as spacing out doses / prolonging the time the drug has an effect on the brain are. I can't comment on your specific doses mentioned, but I suppose they were just guidelines as well. In any case, the duration of the drug has just as much to do with neurotoxicity as dose level does.
