Methods for idenitfying novel alkaloids

What method is used to identify alkaloids in plants? With GC/MS they need a reference chart correct? If they don't have the reference chart how is it one identifies the structure of a novel alkaloid in plants?

Combination of GC/MS, NMR, lots and lots of synthesis, and maybe some polarimetry.
