Mechanism by which stimulants raise seizure threshold and cause mortality

Many stimulants which in particular modulate the dopamine system raise the seizure threshold. For example, aminotetralin causes seizures at 100mg.

What is the mechanism by which these types of stimulants raise the seizure threshold.

Also, what are the specific mechanisms of mortality that can be implicated in fatal stimulant overdoses?
 
I think you mean they lower the threshold. It is a D1 effect...D1 receptor activation reduces the seizure threshold.

The mechanism is probably linked to downstream effects of D1 activation on ion channels.

One mechanism of stimulant-induced fatality: hyperactivity, hyperthermia, hypertension, seizures, vasospasm...leading to rhabdomyolysis...leading to organ failure

Would d1 antagonists be a therapeutic area to look into to raise the seizure threshold?

What do you think is the most common cause of, say, amphetamine induced fatality.
