Does anyone here know anything more about dapoxetine than what was in the news? In depth research you can post? Also, does anyone know if what I drew here is the chemical structure of it (it's S-(+)-N,N-dimethyl-a-[2-(naphthalenyloxy)ethyl] benzenemethanamine):

It's an SSRI and it's in trial for premature ejaculation treatment...
 
Don't know what news you read, but apparently its too short acting for use as an anti-depressant, but is being investigated for premature ejaculation, as Bilz0r said.

It was said to increase the length of time that men with premature ejaculation lasted from around 1 minute to around 3.5 minutes..:\
 
specialspack, I know, I could care less about antidepressants, I'm interested in the pharmacology of a drug that treats premature ejaculation hehe. It amazes me that any serotonin reuptake inhibitor can boost sexual enjoyment and performance (the latest clinical trials found it increases PLEASURE, not just the duration of the sex), no matter how selective.

I also really want to know if what I drew is what they're talking about.

This thread, while having a lot of crap in it, has some interesting stuff and references to the sex side effects of SSRIs.

I'd love to get the discussion of how serotonin is involved in the process of orgasm (in males and females) again, as it seems very interesting. Unfortunately it also seems very complex and without much consensus.

Don't know where you got this idea from. More than 50% of patients taking a SSRI for depression quickly develop sexual dysfunction: loss of libido, anorgasmia, erection problems. SSRIs are sex killers.
 
How in the Hell can you have an ejaculation on any SSRI anyway? Seems like a contradictive medicine. .

SSRI's make me I don't care and can't as far as sex goes. Umong other intolerable side-effects
 
SSRIs are well known for helping delay ejaculation. This is the only sexual side effect my husband suffers from (possibly, he is very slightly less hard than otherwise). He takes zoloft, 100mg/day. He has no libido normally, and the zoloft does not seem to affect it one way or the other.

From what I have read, both the 5HT(2A) and 5HT(1A) receptors seem to be involved in orgasm/ejaculation. SSRIs, by stimulating the 5HT(2A) receptor and inhibiting the 5HT(1A) recptor, inhibit ejaculation.

Fortunately the erectile dysfunction that also often occurs with SSRIs seems to be mediated by the NO pathway and thus can be treated independently.
 
C6H6, the clinical studies on dapoxetine have found it increases sexual enjoyment. I am not talking about all SSRI's, I'm talking about this one which seems to be very out of the ordinary. Why is this one a sex enhancer when the ones used as antidepressants are bad for most peoples' sex drives?
 
Couldn't find any clinical study with dapoxetine on Medline.

However, you got to look at the population where it was tried. I've read other studies where SSRI were tried for premature ejaculation treatment. The men in these trial often ejaculate so prematurely that they are "done" already before the penis is even inside the vagina.

Of course for these men (and their partners) just the ability to achieve an intravaginal ejaculation will increase enjoyment and satisfaction with sex. It does not mean that the average man will benefit from the drug, and he might well develop anorgasmia, which is no fun either.

It you want something to improve sex, I'd look at the dopamine D4 partial agonist ABT-724.
